摘　　要：目的 :观察洛莫司汀 (CCNU)控释药膜对动物实验肿瘤的抑制作用及其急性毒性。方法 :CCNU控释膜埋植入荷瘤小鼠体内 ,观察对鼠艾氏实体瘤的抑制作用以及对血清SOD活性的影响;药膜于体外细胞培养液中进行释放 ,释放液同K562细胞共同培养 ,MTT法观察对K562细胞的抑制率 ,通过透射电子显微镜观察对K562细胞凋亡的影响 ;与口服给药方式相比较 ,通过血液指标和病理组织切片观察其急性毒性。结果 :CCNU控释药膜在体内、外均显著抑制肿瘤细胞的生长 ,增加SOD活性 ,并可诱导肿瘤细胞的凋亡发生 ,且毒性较口服给药明显降低。结论 :CCNU控释药膜可以有效地抑制肿瘤生长 。OBJECTIVE:To observe the inhibiting effect and acute toxicity of CCNU(Lumustine) slow-released film on tumor cells in experimental animals METHODS:The slow-released film was implanted into the body of EAC-bearing mice to observe its inhibiting effect on EAC and the influence on serum SOD activity The film was first released in culture medium,then K562 cells were cultured in this medium to observe the inhibiting rate of K562 cells by MTT method and apoptosis of K562 cells by TEM The acute toxicity of CCNU via film application was detected by blood parameters and pathological section in comparison with that via oral administration RESULTS:The film of CCNU could significantly inhibit the proliferation of tumor cells in vivo and in vitro,increase the SOD activity and induce the apoptosis of tumor cells The toxicity was obviously lower compared with oral administration of CCNU CONCLUSION:The slow-released film of CCNU can effectively inhibit the growth of tumor with low toxicity
