摘 要:以新籽瓜2号等籽瓜(Citrullus Lanatus ssp.vulgaris convar.megalaspermus Lin et Chao.)品种为材料,进行了子叶外植体切块组培中成苗因子的研究。结果表明,幼苗子叶是籽瓜组织培养的适宜外植体。MS+2mg/L 6-BA+0.1mg/L IBA培养基是子叶不定芽诱导的适宜培养基,芽伸长培养基以MS+0.2mg/L KT为宜。MS+0.5mg/L IAA是诱导生根的最佳培养基。试验初步建立了适用于籽瓜遗传转化的子叶快速高频再生系统。Seed watermelon ( Citrullus lanatus ssp. vulgaris convar. megalaspermus Lin et Chao.) is the one convar. of ssp. vulgaris. It is one of the well-known, high quality seed-use watermelon. This paper reports how the factors affecting tissue culture as we aims to establish a quick, high frequency regeneration system of genetic translation for seed watermelon. The results indicated that the basal and middle segments of cotyledon of the young plants of 1 to 6 days age were the best appropriate explants. The combination of 6-BA 2 mg/L+IBA 0.1 mg/L was the optimum medium for differentiation of buds. The adventitious buds were found at the basal parts of central or lateral veins. The adventitious shoot elongation was the best on MS supplemented with 0.2 mg/L KT. On MS plus IAA 0.5 mg/L medium, the shoots stretched better and the rooting rates was 80%. This experiment has explored the suitable culture medium for bud inducing, shoot elongating and rooting which fit cotyledon. Through the test and verification at the National Key Biotechnology Laboratory for Tropical Crops, the results were proved to be steady and reliable, it is believed that the research will be useful for the seed watermelon tissue culture, utiliazation of so-.matic clonal variation and its genetic transformation.
