摘 要:目的 观察大鼠放射诱导脑损伤后海马区IL - 6mRNA的动态表达 ,探讨其在放射性脑损伤急性期反应的发病机制中可能的作用。方法 制备大鼠脑放射性损伤模型。应用逆转录聚合酶链反应 (RT -PCR)半定量分析大鼠脑放射性损伤后海马区在不同时间、不同剂量水平IL - 6基因转录的动态表达。结果 正常组海马区IL - 6mRNA低水平表达 ;全脑照射后表达逐步上调并在 12h达峰值 ,是正常组的 2~ 5倍 (P <0 .0 5 ) ,15Gy组上升幅度最大 (P <0 .0 0 1) ,高出 2Gy组和 30Gy组 1倍多 (P <0 .0 5 ) ;30Gy组低于 2Gy组 ,但无显著差异 (P >0 .0 5 ) ;在 6h时间点也可见到这一趋势 ,15Gy组与 30Gy组仍有显著差异 (P <0 .0 5 )。各照射组在 2 4h后恢复到基础水平。 结论 大鼠放射性脑损伤后海马区IL - 6mRNA表达上调 ,参与了脑放射性损伤急性期的细胞反应。Objective To discuss the possible role of interleukin-6(IL-6) in the mechanism of acute phage of brain radiation injuries by referring to observation of the expression of the cytokine in the rat hippocampus. Methods A model of brain radiation-induced injuries in the rat was deve- loped. Semiquantitative analysis of IL-6 in the hippocampus was done at different time and dose level after whole-brain irradiation with reverse-transcription polymerase chain reaction (RT-PCR). Results The expression of IL-6 in the hippocampus was at lower level in normal group; It was up-regulated gradually after whole-brain irradiation and reached the peak level in 12 h,which was 2~5 times as that in the normal group( P <0.05); Its radiation-induced level in 15 Gy group was the highest( P <0.001), one time higher than that in the 2 Gy group and 30 Gy group( P <0.05); The increased level in the 30 Gy group was lower than that in the 2 Gy group, but there wasn′t significant difference. The tendency in 6 h was also observed. There was significant difference between the 15 and 30 Gy group( P <0.05). The expression in irradiated groups returned to the basic level in 24 h. Conclusion The expression of IL-6 in the rat hippocampus after radiation induced injuries is up re- gulated and shows that the cytokine may play a role in the cells reaction in the acute phase of brain radiation-induced injuries.
