摘　　要：目的:构建鼠LTC4合酶基因靶向敲除载体,用于转染鼠胚胎干细胞,建立LTC4合酶基因敲除鼠。方法:分离、克隆LTC4合酶基因的1.85kb和1.35kb片段,作为靶向载体的5'同源臂和3'同源臂,重组到pJNS2载体中,筛选鉴定阳性重组子。结果:构建的靶向载体由5'同源臂-Neo-3'同源臂-hTk-pJNS2构成。结论:经DNA测序及限制性酶切鉴定分析,该构建载体结构及序列正确,可用于转化鼠胚胎干细胞。Objective:A targeting vector was constructed for disrupting LTC 4 synthase in ES of mouse.Method:1.85kb and1.35kb fragments of LTC 4 S were isolated and cloned to use as the5'homologous arm and3'homologous arm of the targeting vector.Inserting5' homologous arm and3' homologous arm into pJNS2generated recombinant of pJNS2.The positive recombinants were screened and identified.Result:The targeting vector consisted of5'homologous arm-neo cassette-3'homologous arm-hTk-pJNS2.Conclusion:The structure and sequence proved to be correct with DNA sequencing and restriction endonuclease digestion analysis.
