摘  要:Aim To purify and characterize flammulin, a basic protein with anti-tumoractivities. Methods Ammonium sulfate, ethanol fractionation and column chromatography were used forseparation and purification. Electrophoretic analysis, amino acid analysis, and MS of flammulin werecarried out. Results Flammulin was purified to electrophoretic homogeneity and crystallized. With amolecular mass of 19891.13 Da, pI 8.9, λ_(max) = 276 - 278 nm, λ_(min) = 250 nm, flammulin wascharacterized by its lack of methionine. Fingerprint mapping of flammulin was determined by MALDI-MSfollowing in-gel protease digestion; no close matches were identified. Conclusion Flammulin waspurified to electrophoretic homogeneity, and its characteristics are discussed for the first time.目的 对一种具有抑瘤活性的碱性蛋白火菇素进行分离纯化和性质研究。方法 应用硫酸铵、乙醇分级沉淀 ,柱色谱等方法进行分离纯化。电泳分析、氨基酸组成分析、质谱分析等方法被用于火菇素的理化性质研究。结果 火菇素被提纯至电泳纯并结晶。经性质研究确定火菇素的分子量为 19891.13Da ,pI=8.9,λmax=2 76~ 2 78nm ,λmin=2 5 0nm ,并缺少甲硫氨酸。通过基质辅助激光解吸离子化质谱 (MALDI—MS)分析 ,测得了火菇素的指纹图谱 ,表明它为一种新蛋白。结论 火菇素被首次提纯至电泳纯 。
