摘  要:目的 :了解灭活SARS病毒肌肉注射途径对系统免疫的影响。方法 :将 5周龄雌性BALB/c小鼠随机分为两组 ,每组 1 0只 ,免疫组小鼠每只注射 5 0 μg的灭活病毒悬液 ,对照组注射等体积PBS。每组分别在免疫的 8d和1 4d处死 5只小鼠 ,分离脾细胞和外周血淋巴细胞 ,流式细胞仪观察其淋巴细胞表型。结果 :第 8天脾脏的淋巴细胞表型变化较为明显 ,CD+ 4 T细胞的构成比增加 (P =0 .0 0 1 ) ,CD+ 4 /CD+ 4 比值明显增多 (P =0 .0 0 7)。外周血淋巴细胞表型在第 1 4天变化明显 ,与对照组有极显著差异 ,CD+ 4 ,CD+ 8,CD+ 3 T细胞构成比和T/B比值显著降低 (P =0 .0 0 5 ,0 .0 0 0 ,0 .0 0 6和 0 .0 0 1 ,) ,同时CD4 5R/B2 2 0 + 细胞 (B细胞 )构成比显著增加 (P =0 .0 0 0 )。结论 :肌肉途径免疫灭活SARS病毒早期即可诱导机体系统免疫细胞表型的变化 。Objective:To understand the immune mechanism of the β_propiolactone inactivated SARS virus in BALB/c mice.Methods:Five_week_old female BALB/c mice were immunized with a single dose of 50?μg of the inactivated virus by intramuscular injection. The control mice were given equal volume of PBS. Five mice of each group were sacrificed on days 8 and 14, and lymphocytes from the spleen and peripheral blood were isolated. The lymphocyte phenotypes were detected by FACS. Results:The splenocyte CD + 4 T cells increased significantly on day 8 ( P =0.001); the ratio of the CD + 4 to the CD + 8 T cells increased significantly on day 8th( P =0.007). The peripheral blood lymphocyte phenotype changed little on day 8, but on day 14 the phenotype changed significantly in the immunized mice. The CD + 4, CD + 8, CD + 3 T cells and the ratio of the T cells to B cells significantly decreased than the control ones( P =0.005, 0.000, 0.006 and 0.001, respectively); whereas the CD45R/B220 + cells (B cells) increased significantly ( P =0.000). Conclusions:The inactivated SARS virus immunized by intramuscular injection could induce systemic immune response in BALB/c mice.
