Effect of sublethal doses of abamectin and β-cypermethrin on glutathione S-transferases in diamondback moth Plutella xylostella(L.)

摘  要:研究了阿维菌素和高效氯氰菊酯亚致死剂量对小菜蛾Plutellaxylostella(L .)谷胱甘肽S -转移酶 (GSTs)活性的影响。用亚致死剂量的阿维菌素和高效氯氰菊酯处理小菜蛾敏感品系后 ,二处理组的GSTs活性比对照组分别增长了 4 2 %和 70 % ;抗性品系经上述 2种药剂同样处理后的活性比对照分别降低了 4 5 %和 30 %。对GSTs的动力学研究表明 ,敏感品系用高效氯氰菊酯处理后GSTs的Km 值比对照降低了近 4 0 % ,而阿维菌素处理后其Km 值变化不大 ,说明高效氯氰菊酯处理后 ,GSTs对底物的亲和力明显增强 ;抗性品系中二处理组GSTs的KmThe treatment of the sublethal doses of abamectin (AVM) and β cypermethrin (CPM) increased the specific activities of glutathione S-transferases (GSTs) in the susceptible strain (XH-S) of diamondback moth (DBM), Plutella xylostella(L.) by 42% and 70%, respectively. However, the GSTs activities decreased by 45% and 30%, respectively, in the abamectin-resistant strain AV-R with the same treatments. On the kinetic study, we found that with the treatments of AVM and CPM, the affinity of GSTs to 1 chloro 2,4 dinitrobenzene (CDNB) at their sublethal concentrations was varied. The K m(Michaelis Menten constant) value of GSTs decreased by about 40% in XH-S strain treated with CPM compared with that of the control, but there was no significant change in the case of AVM treatment. Moreover significant differences of the K mvalues were not observed in the AV-R strain with or without the treatment of AVM and CPM.
