芬太尼对小鼠脑内摄取^(125)I-β-CIT的影响

摘  要:目的 探讨阿片受体激动剂芬太尼对小鼠脑内摄取1 2 5I 2 β 甲酯基 3β (4 碘苯基 )托烷(1 2 5I β CIT)的影响。 方法  6 5只昆明小鼠注射不同剂量芬太尼 ,观察其对小鼠脑内摄取1 2 5I β CIT的影响 ;比较芬太尼注射后间隔 10min与 1h1 2 5I β CIT在脑内的摄取。 结果 芬太尼注射剂量为按体重 12 5~ 30 0 μg kg时 ,纹状体对1 2 5I β CIT的摄取明显增多 ,且海马、小脑的每克组织百分注射剂量率(%ID g)及全脑摄取量 (%ID)均高于对照组 ;而剂量为按体重 12 5~ 10 0 μg kg时 ,纹状体 %ID g均比对照组低 ,且额叶皮质、海马 (除 6 2 .5和 12 .5 μg kg组 )、脑干 (除 6 2 .5 μg kg组 )、小脑的 %ID g及全脑 %ID均低于对照组。按体重注射芬太尼 12 5 μg kg后间隔 10min再注射1 2 5I β CIT ,小鼠的纹状体、额叶皮质、海马、脑干、小脑 %ID g ,全脑 %ID及纹状体特异性摄取率 (T CB 1)均高于间隔 1h注射1 2 5I β CIT组。按体重注射芬太尼 5 0 μg kg ,于1 2 5I β CIT注射后 1h取脑标本 ,则小鼠纹状体、额叶皮质、海马、脑干、小脑 %ID g及全脑 %ID均高于对照组 ,但差异无显著性 ,且T CB 1亦高于对照组。结论 芬太尼在不同剂量和时间对脑内摄取1 2 5I β CIT的影响不同。Objective To investigate the effect of fentanyl on 125 I 2β carbomethoxy 3β (4 iodophenyl) tropane ( 125 I β CIT) uptake in mice brain. Methods ①KM mice groups of five were given different doses of fentanyl, and 10 min or 1 h later were given a dose of 125 I β CIT. ②Two groups of animals were killed at 2 h after injection of 125 I β CIT. ③One group of animals were killed at 1 h after injection of 125 I β CIT. Results ①In the striatum, frontal cortex, hippocampus, brain stem, cerebellum and whole brain, a dose dependent increase in uptake (%ID/g or %ID) of 125 I β CIT was detected at the fentanyl doses ranging from 125 to 300 μg/kg, and the uptakes of hippocampus and cerebellum were higher than that of the controls. There was a great difference in the value of %ID/g or %ID between the group treated with 250 μg/kg fentanyl and the control group; while at the doses from 12.5 to 100 μg/kg, a dose dependent decrease in uptake in the same regions was observed and all the uptake levels were lower (hippocampus: except 62.5 and 12.5 μg/kg groups; brain stem:except 62.5 μg/kg group) than that of the controls. ②The uptakes of 125 I β CIT in the striatum, frontal cortex, hippocampus, brain stem, cerebellum and whole brain in the groups injected with 125 I β CIT 10 min after fentanyl treatment were higher than that in the groups injected with 125 I β CIT 1 h after fentanyl treatment. ③The binding of 125 I β CIT in the striatum, frontal cortex, hippocampus, brain stem, cerebellum and whole brain in the groups killed at 1 h after injection of 125 I β CIT was higher than that in the control group, but without significant difference. Conclusion Fentanyl may have different effects on 125 I β CIT at various time points and doses.
