摘　　要：目的 :观察牛膝醇提物 (Nx E)对Aβ42 聚集的抑制作用。方法 :Aβ42 单独或和Nx E在 37℃ ,5 %CO2 培养箱中孵育后 ,采用硫磺素T(Th T)荧光分析以及透射电镜观察Aβ42 的聚集程度。结果 :1μg·μl 1Aβ42 孵育 7d后荧光强度明显升高 ,电镜观察发现Aβ42 以淀粉样纤维结构为主 ,呈针状聚集 ;低、中、高三个剂量 (7 5、15、30 μg·μl 1)的Nx E与Aβ42 共同孵育 7d后 ,中、高剂量Nx E与Aβ42 的共同孵育产物荧光强度比Aβ42 单独孵育明显减弱 ;形态学观察纤维样结构减少 ,无定形结构增加 ,存在大量散在的颗粒。结论 :1μg·μl 1Aβ42 孵育 7d已“老化” ;15和 30 μg·μl 1的Nx E能在一定程度上抑制Aβ42 聚集。Objective: To observe inhibiting effect of ethanol extract from Achyranthes bidentata (Nx E) on Aβ 42 (β amyloid protein 42) aggregation and fibril formation Methods: Aβ 42 or Aβ 42 and Nx E were incubated in 37℃,5% CO 2 incubator first, fibril formation was monitored by thioflavine T fluorescence and was observed with transmission electron microscopy Results: At 7 days, 1 μg·μL 1 Aβ 42 fluorescence intensity was significantly increased, and much amyloid like fibril was observed Aβ 42 co incubated with 15 and 30 μg·μL 1 Nx E for 7 days, their fluorescence intensities were remarkably reduced, few fibrils but an amorphous material were often observed Conclusions: After 1 μg·μL 1 Aβ 42 was incubated alone up to 7 days, Aβ 42 aggregated and amyloid like fibril formed 15 and 30 μg·μL 1 Nx E can prevent Aβ 42 from polymerizing to some extent
