摘 要:目的探讨中药强骨类方剂对抗失重骨丢失的作用机制,及对尾吊大鼠甲状旁腺及肾组织的影响。方法用50只SD大鼠尾吊-30°21d模拟失重,观察强骨抗萎方对大鼠肾线粒体膜流动性、过氧化脂质(LPO)、超氧化物歧化酶(SOD)、Na-K-ATP酶和腓肠肌肌酸磷酸激酶(CPK)活性的影响,并对其甲状腺进行形态学观察。结果与正常对照组比较,尾吊组大鼠肾线粒体膜偏振度、微粘度和LPO均有升高趋势,SOD无明显变化;肾Na-K-ATP酶和腓肠肌CPK活性均有增加趋势。与尾吊模型组比较,中药复方小剂量组肾线粒体膜偏振度、微粘度均有降低趋势;大剂量组LPO明显降低(P<0.05);大中小各剂量组肾组织SOD、Na-K-ATP酶、中小剂量组腓肠肌CPK活性均明显降低(P<0.01或P<0.05);尾吊模型组大鼠甲状腺特别是甲状旁腺处于较强的分泌功能状态,中药复方小剂量可使甲状腺功能状态接近正常。结论强骨抗萎方可调节尾吊大鼠甲状旁腺和肾组织的功能,维持骨和血清钙的平衡,从而有效对抗失重骨丢失。Aim To investigate the mechanism of Qianggu Kangwei compounds against b one loss in the weightlessness state and its effects on parathyroid glands and k idney in tail-suspended rats.Methods Fifty SD rats were tail-suspended (-30° ) for 21 days to simulate the state of weightlessness.The effects of this compou nd on liquidity of mitochondrial membrane fluidity,lipide peroxide(LPO),activiti es of superoxide dismutase(SOD),Na-K-ATPase in kidney and creatine phosphokina sein(CPK) in gastrocnemius muscle in these rats were examined, and thyroid gland was observe morphologically.Results Compared with the control group,the polariz ation range of mitochondrial membrane,LPO,activities of Na-K-ATPase in kidney and CPK in gestrocnemius muscle had an increased tendency in the rats with tail suspended, with no significant changes of SOD in activity.Compared with tail-su spended group, the polarization range of mitochondrial membrane,LPO,activities o f Na-K-ATPase in kidney and CPK in gestrocnemius muscle had a decreased tenden cy in the TCM group(P< 0.05-0.01), with the activity of LPO decreased significa ntly in the large-dosage group(P< 0.05).The rats with tail suspended were in a strong secretory function status of parathyroid glands. The small dosage group s howed the effect in inhibition of the abnormal function of parathyroid glands in suspended rats and made it normalization. Conclusion Qianggu Kangwei compound r egulates the function of parathyroid glands and kidney, and keep the balance of calcium between serum calcium and bone. Therefore, it is effective against bone loss in weightlessness.
