摘 要:目的探讨氟对体外培养长骨生长发育、形态结构和骨组织生化指标的影响。方法采用体外骨器官培养实验方法,观察不同剂量氟(0.0、2.5、5.0、10.0、20.0μg/ml)对小鼠胚胎长骨生长发育、形态结构和骨组织碱性磷酸酶(AKP)、羟脯氨酸(Hyp)、脂质过氧化物(LPO)、超氧化物歧化酶(SOD)等生化指标的影响。结果染氟2.5μg/ml时,尺骨生长发育、形态结构与对照组差异无显著性(P>0.05),但有刺激长骨生长的趋势;骨组织Hyp和LPO含量与对照组差异亦无显著性(P>0.05),但SOD活性明显低于对照组(P<0.05),而AKP活性明显高于对照组(P<0.05)。当染氟剂量≥5.0μg/ml,随着染毒剂量的增加,尺骨和尺骨骨干生长速度明显减慢,尺骨面积和尺骨骨干面积明显减少(P<0.05,P<0.01),形态结构出现异常改变;骨组织Hyp、LPO含量显著升高(P<0.05,P<0.01),骨组织SOD活性进一步明显降低(P<0.01)。染氟10.0μg/ml和20.0μg/ml,AKP活性明显降低(P<0.05,P<0.01),但染氟5.0μg/ml时,骨组织AKP活性与对照组差异无显著性(P>0.05)。结论过量氟对体外培养长骨生长发育、形态结构和骨Hyp、LPO含量以及骨AKP、SOD活性有明显的不利影响。Objective To study the in vitro effects induced by fluoride on the growth and development,morphological struc-ture and biochemistry indices of long bone.Methods Mouse fetal ulnas exposed to0.0,2.5,5.0,10.0and20.0μg/ml fluoride were cultured in vitro.The growth and development,morphological changes of the cultured bone were observed and the contents of hydroxyproline(Hyp)and lipid peroxidation(LPO),and the activities of alkaline phosphatase(AKP)and superoxide dis-mutase(SOD)in the bone tissues were determined.Results At the2.5μg/ml fluoride level,the morphological structure,growth and development of the bone were not significantly different from those of the controls(P>0.05),but there was a tenden-cy for growth stimulation.Hyp and LPO contents of the bone tissue were not markedly different from those in the control group(P>0.05),while the activities of SOD of bone tissue were lower(P<0.05)and the activities of AKP were significantly higher(P<0.05)than those of the control group.When the fluoride concentrations were≥5.0μg/ml,the ulnar growth and develop-ment were retarded,the ulnar length and area,diaphysial length and area decreased significantly with the increase of fluoride concentration(P<0.05,P<0.01),and the morphological structure of the bone appeared obviously abnormal.Hyp and LPO con-tents increased significantly with the increased concentration of fluoride(P<0.05,P<0.01).The bone SOD activities decreased significantly(P<0.01).In the5.0μg/ml fluoride group the activity of AKP decreased,but it was not significantly different from that in the control group.At 10.0-20.0μg/ml fluoride the activities of AKP decreased markedly with the increase of fluoride concentration.Conclusion It showed that excess fluoride exposure could have adverse effects on the growth and development,morphological structure,the contents of Hyp and LPO,and the activities of AKP and SOD of cultured long bone.
