摘 要:目的 探讨慢性染铅大鼠海马脑源性神经营养因子(Brain derived neurotrophic factor,BDNF)的变化。方法 Wistar大鼠分别饮用0.02%、0.2%的醋酸铅(PbAc)溶液,染铅3个月后,通过Y-迷宫实验检测各组动物学习记忆行为变化,采用免疫组织化学ABC法观察各组大鼠海马CA1区、CA3区和齿状回BDNF阳性神经元数目的改变。结果 在正常情况下,BDNF样免疫阳性神经元在海马各区及齿状回均有分布。与对照组相比,饮用0.02%和0.2% PbAc两组染铅大鼠学习记忆能力明显降低(P<0.05),同时两组染铅大鼠海马CA1区、CA3区和齿状回的BDNF阳性神经元数均明显减少(P<0.05)。结论 海马各区BDNF阳性神经元数目的减少可能是大鼠染铅后学习记忆功能受损的机制之一。Objective To investigate the changes in brain-derived neurotrophic factor( BDNF) in hippocampus of chronic lead-exposed rats.Methods Wistar rats were exposed to lead by drinking 0.02% and 0.2% lead acetate solution,separately,for three months/Hie chan-gesin ability of learning and memory were investigated through Y-labyrinth experiment,and the changes in BDNF positive neuron in CA1, CA3 and dentate gyms in hippocampus of rats were observed by immunohistochemistry. Results BDNF positive neurons distribute every where in the hippocampus in normol condition. Compared with the control group, the BDNF positive neurons in the hippocampus were decreased in number significantly in the two different dosage (0.02% ,0.2%)chronic lead-exposed rats ( P < 0.05). Conclusion The reduction of BDNF positive neuron in hippocampas might be related to the impact of lead on learning and memory in rats.
