摘 要:目的探讨丹参对顺铂耳蜗毒性中细胞凋亡的防护作用。方法选取18只耳廓反射灵敏的豚鼠分为正常对照组、顺铂组和丹参组,每组6只。运用ABR、电镜和TUNEL等技术行各组动物听功能和耳蜗毛细胞、螺旋神经节细胞凋亡的测试。结果顺铂组和丹参组动物用药后ABR阈值(44.58±6.89 dB,22.50±4.37 dB)与用药前(13.33±3.46 dB,13.46±2.91 dB)比较有明显升高(P均<0.01)。电镜和TUNEL均见到耳蜗毛细胞、螺旋神经节细胞和血管纹细胞出现凋亡细胞,而正常对照组动物无凋亡现象。顺铂组ABR阈值和TUNEL阳性细胞数均明显高于丹参组(P均<0.01)。结论丹参能够抑制顺铂诱导的耳蜗毛细胞、螺旋神经节细胞等细胞的凋亡,改善听功能。Purpose To explore the protective effect of salvia miltiorrhiza on the cisplatin-induced apoptosis in cochlea of guinea pigs. Methods Eighteen guinea pigs were divided into three groups: cisplatin group, salvia miltiorrhiza group and control group. There were six guinea pigs in each group. The andition of animals smd the. hair cell, ganglion spiral cell apoptosis in cochlea were tested with ABR, electron microscopic and TUNEL technology. Results The ABR threshold of cisplatin group and salvia miltiorrhiza group (44. 58 ± 6. 89 dB, 22. 50 ± 4. 37 dB) after being applied with drug were apparently higher than those (13.3 ± 3.46dB, 13.46 ± 2.91 dB) before being applied with drug( P < 0.01) . Apoptosis was happened in hair cell, ganglion spiral cell and stria vascularis cell in the cochlea of guinea pigs after being tested with electron microscopic and TUNEL technology. But there was no apoptosis in control group. The ABR threshold and TUNEL positive number of cisplatin group were apparently higher than those of salvia miltiorrhiza group( P < 0. 01) . Conclusion Salvia miltiorrhiza can inhibit the cisplatin-induced apoptosis of hair cell, ganglion spiral cell etc. in the cochlea of guinea pig, improve the auditory function.
