摘  要:目的:观察脑醒喷鼻剂对大鼠局灶性脑缺血-再灌注损伤脑组织自由基及一氧化氮合成酶(NOS)变化的影响。方法:阻断大鼠左侧大脑中动脉,建立大鼠MCAO局灶脑缺血再灌注模型,用比色法检测丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)及一氧化氮合成酶(NOS)。结果:高剂量脑醒喷鼻剂使MDA生成减少,SOD含量升高,增加GSH-PX与NOS活性明显,与尼莫通对照组比较P>0.05,与生理盐水组比较P<0.05。结论:脑醒喷鼻剂能防止脑缺血缺氧所致的脂质过氧化,清除自由基损害,并增加内皮型NOS(eNOS)活性,对脑组织的缺血再灌注损伤有保护作用。Objective:To observe the effects of Naoxing nasal spray (NXNS) on free radical and nitric oxide synthase (NOS) of brain tissues in rats with focal cerebral ischemia reperfusion injury (CIRI). Methods:Rat model of cerebral ischemia/reperfusion was established by the use of left middle cerebral artery occlusion (LMCAO). We measured the free radical (MDA, SOD, GSH-PX) and NOS by using chromatometry after CIRI. Results:In high dosage of NXNS group, the cerebral levels of MDA was reduced and that of SOD was increased. The activities of GSH-PX and that of NOS were elevated. The differences were significant compared with normal saline group (P<0.05), while not significant compared with Nimodipine group (P>0.05). Conclusion:It is indicated that Naoxing nasal spray can prevent the damage of lipid peroxidation from cerebral ischemia, clean lesion of free radical and increase the activity of endothelium NOS, therefore it could protect cerebral tissues from CIRI.
