庚型肝炎病毒“重庆株”NS3区核苷酸和氨基酸序列分析

摘  要:目的 :研究庚型肝炎病毒 (HGV)“重庆株”(CHq)在NS3区与其它一些分离株之间核苷酸和氨基酸序列的同源性。方法 :采用RT -套式PCR ,从中国重庆市的庚型肝炎患者血清中分离出HGV -NS3区基因cDNA并测序 ,然后经计算机软件对其核苷酸序列及由此推导的氨基酸序列进行分析。结果 :HGV“重庆株”NS3区与HGV - 1(U4440 2 )、HGV - 2 (U45966)、GBV -C(U6371 5)及HGVC964NS3区之间 ,核苷酸序列同源性分别为 85 .96 %、83 .67%、85 .96 %和 96 .2 8% ;氨基酸序列同源性分别为 97.4 %、97.4 %、97.4 %和 96 .6 %。结论 :在NS3区核苷酸序列方面 ,“CHq”与HGVC964同源性最高 ,而在NS3区氨基酸序列方面 ,“CHq”与其它 4株HGV的同源性均在 96 %以上 ,无明显差别。提示以NS3蛋白为抗原 ,对感染不同HGV株的患者血清进行免疫学检测 ,其检出率应无显著差异。Objectives: To determine the homology of the nucleotide and amino acid sequences between the hepatitis virus G (HGV) Chongqing strain (CHq) and some other strains derived from different countries or regions. Methods: A HGV NS3 gene cDNA was amplified from a patient with hepatitis G in Chongqing City by RT-nested PCR, then followed by nucleotide sequencing and analyzed the amino acid sequence with computer programs. Results: In the NS3 region, between HGV 'CHq' and HGV-1 (U44402),HGV-2(U45966),GBV-C(U63715) and HGV C964, the identities of the nucleotide sequences were 85.96%,83.67%,85.96%,and 96.28%,respectively,and the identities of amino acid sequences were 97.4%,97.4%,97.4% and 96.6%, respectively.Conclusions: 'CHq' is closer to HGV C964 in nucleotide sequence. However, there is no significant difference between different HGV strains in the putative amino acid sequences, that is, with HGV NS3 protein or peptide as the antigen, the positive anti-HGV NS3 detectable rate in the sera from the patients infected with different HGV strains might be similar.atientwithhepatitisGinC
