摘 要:目的 探讨白细胞介素 -1β( IL -1β)在体外受精 -胚胎移植 ( IVF-ET)患者卵巢黄素化颗粒细胞表面的表达及与生殖内分泌的关系。 方法 3 6名 IVF-ET妇女 ,在采用长周期促超排卵方案 ,注射人绒毛膜促性腺激素 ( h CG) 1 0 0 0 0 IU后 3 4~ 3 6h超声引导下经阴道取卵时留取卵泡液 ,离心后分离黄素化颗粒细胞 ,应用流式细胞计 ( FCM)检测细胞表面 IL-1 β的表达 ,用酶联免疫吸附法 ( EL ISA)检测卵泡液中 IL-1 β浓度 ,用放射免疫法检测卵泡液和血清中激素浓度。 结果 行 IVF-ET激素刺激周期妇女的卵泡液中可检测到 IL -1β,血清 IL -1β浓度类似 L H的变化规律。黄素化颗粒细胞表面有 IL -1β表达。h CG注射日血清 IL-1 β浓度与雌二醇 ( E2 )浓度呈正相关 ( r=0 .92 ,P<0 .0 0 1 ) ,与孕酮 ( P)呈负相关 ( r=-0 .3 4,P<0 .0 5)。 结论 本研究在人黄素化颗粒细胞表面检测到 IL -1β蛋白表达 ,说明卵泡液中 IL -1β不仅来源于血液 ,而且也由黄素化颗粒细胞分泌。提示 IL-1 β可能通过自或旁分泌方式参与生殖内分泌活动。Objective: To detect the expression of interleukin 1β (IL 1β) on the surface of human luteinizing granulosa cells and the relationship between IL 1β level and reproductive endocrinologic hormone in human follicular fluid and sera. Methods: 36 women with age (30.1±2.5) years old, body mass index (BMI)=(20.1±3.2) kg/m 2 undergoing in vitro fertilization embryo transfer (IVF ET) from Nanfang Hospital were evaluated.Human luteinizing granulosa cells were recovered from follicular fluid obtained during transvaginal ultrasound guided oocyte retrieval for IVF ET. Flow cytometry(FCM) was used to detect the expression of IL 1 β on the surface of human granulosa cells. The concentrations of IL 1β and hormone in human follicular fluid and sera were measured by enzyme linked immunosorbent assay (ELISA) and radioimmunoassay (RIA) respectively. Results: IL 1β was detected in follicular fluid of IVT ET women and on the surface of human luteinizing granulosa cells. A positive correlation was observed between the sera concentrations of IL 1β and E 2 on the day of injecting hCG( r=0.92,P<0.001 ). The concentration of IL 1β was negatively correlated with the progesterone level on the day of hCG injection ( r=-0.34,P<0.05 ). Conclusion: These data suggest that the IL 1β in the follicular fluid comes from not only plasma super filtrate but also luteinizing granulosa cells. IL 1 β might participate in reproductive endocrinologic activity through autocrine and paracrine.
