摘　　要：我们研究了人骨形成蛋白-2复制物(recombinant human bone morphogenetie protein-2,rhBMP-2)及其载体对MC3T3-E1细胞的作用。观察到使用羟基磷灰石(hydroxyapatite,HAP)载体比玻璃或钛载体获得了较高的碱性磷酸酶(alkaline phosphatase ALP)活性。当胶原被加入到HAP载体上时,ALP活性和DNA含量极其显著地增加。当细胞在包被胶原的HAP载体上培养而不是在HAP载体上时,rhBMP-2增加ALP活性。这些结果暗示rhBMP-2刺激MC3T3-E1细胞分化,以及包被胶原的HAP对BMP是很有用的载体。We examined the effects of recombinant human bone morpbogenetic protein-2(rhBMP-2) and its carriers on osteoblastic MC3T3-E1 cells.Much higher alkaline phosphatase(ALP) activity was observed with the hydroxyapatite ( HAP ) carrier than with the titanium or glass carrier.The ALP activity and DNA content increased remarkably when collagen was added to the HAP carrier.The rhBMP-2 increased the ALP activity when the cells were cultured on the collagen-coated HAP carrier but not HAP carrier.These findings imply that the rhBMP-2 stimulates the differetiation in MC3T3-E1 cells and the collagen-coated HAP is an effective carrier for BMP.
