摘  要:目的 观察灌胃 (ig)葡萄籽原花青素提取物 (GSPE)对顺铂 (CP)肾毒性作用的防护及可能机制。方法 大鼠经igGSPE 4 5 0mg/kg后 1h ,腹腔注射 (ip)CP 6mg/kg ,给药后第 1,3,5天处死动物。结果 CP组ip后第 3,5天肾脏系数升高 ,肾皮质丙二醛水平与血尿素氮水平升高 ,肾皮质线粒体超氧化物歧化酶活力及谷胱甘肽过氧化物酶活力下降 ,与生理盐水 (NS)组比较 ,差异有显著或非常显著性 (P <0 0 5或P <0 0 1)。GSPE预处理可显著改善CP处理后第 3,5天出现的上述改变。另外 ,单独GSPE组的上述观察指标均接近NS组。结论 GSPE经口给予能明显预防CP所致的肾毒性。其机制可能是GSPE使CP在体内产生的自由基减少 ,脂质过氧化程度减轻 ,提高机体抗氧化能力。Objective To study the protective effect of grape seed proanthocyanidin extract(GSPE)on nephro toxicity induced by cisplatin(CP)in rats.Methods Male SD rats were randomly divided into four groups:namely control,CP (ip 6 mg/kg),CP+450 mg/kg GSPE(GSPE ig 1h prior to CP ip)and GSPE (ig 450 mg/kg GSPE).On 1,3, 5 d after CP treatment,blood and kidney samples were collected.Results The significant nephro toxicity appeared on the 3 rd and the 5 th d after administration in rats.GSPE pretreatment(ig)would alleviate the increase of kidney index,blood urea nitrogen,formation of malondialdehyde in kidney cortex homogenate,and the decrease of activities of superoxide dismutase and glutathione peroxidase in mitochondria of kidney cortex induced by CP in rats.Conclusions The results indicate that GSPE would prevent kidney from the damage induced by CP,and its mechanism might be the inhibition of lipid peroxidation and the enhancement of antioxidation.
