摘  要:目的 :观察雌激素替代治疗后雌性去势大鼠大脑皮质神经元形态学变化 ,探讨雌激素对中枢神经系统的有益作用。方法 :将 9月龄去势雌性SD大鼠 2 0只随机分为用药组 (去势后给予倍美力溶液 0 .2mg/kg·d- 1灌胃 )和对照组 (去势后予以生理盐水灌胃 )。 12周后测定两组血清E2 水平 ,同时对大脑额叶皮质神经元行形态学检查。结果 :(1)用药组E2 水平、脑重 /体重指数及额叶皮质神经元计数均高于对照组 (P <0 .0 1) ;用药组额叶皮质神经细胞凋亡百分率低于对照组 (P <0 .0 1)。 (2 )对照组神经元细胞超微结构显示细胞有老化、凋亡趋势 ,而用药组形态及结构基本正常 ;用药组神经元突触密度较对照组增高 ,突触前膜含神经递质的小泡较对照组多。结论 :去势大鼠应用雌激素替代治疗可延缓神经元衰老 。Objective:To explore the benefits of estrogen on central nerve system by observing morphological changes of neurons in castratived rats after estrogen replacement therapy.Methods:Twenty female SD rats aged 9 months were divided randomly into 2 groups:experimental group(castrated and received conjugated estrogen 0.2mg/kg·d -1 ) and control group(castrated and received 0.9% NS).12 weeks later,serum E 2 levels were determined, and morphological changes of neurons in pallium were observed.Results:(1)Experimental group had higher serum E 2 level and brain/body weight index than those of control group.(2)Experimental group had more neurons count in the brain and a significantly lower percentage of neuronal apoptosis than those in control group.(3)Under electron microscope,the neurons of control group were found to have a trend of aging and apoptosis,while the neuronal conformation and microstructure of experimental group were observed to be normal in substance.It was also found that neurons of experimental group had a higher synapse density and more neural transmitter in synapses than those of control group.Conclusion:Estrogen replacement therapy has benefit on delaying degeneration and aging of central neural cells in castrated female rats.HRT can also reduce the apoptosis and lose of neural cells.
