摘  要:目的 :观察自体骨骼肌干细胞 -卫星细胞移植于犬缺血心肌后对犬输出量及每搏量的影响。方法 :取 12只成年犬臀大肌 ,分离干细跑并培养、传代、标记 ,将DAPI标记的干细胞自左冠状动脉前降支灌注入已经建立的急性心肌梗死动物模型的心肌中。 2 ,4 ,8周后对犬心输出量及每搏量进行观察。结果 :干细胞移植 2 ,4 ,8周后 ,实验组犬心输出量及每搏量与对照组相比均有显著差异 ,以 8周时最为明显 ,具有统计学意义。干细胞移植后的不同时间段里 ,不同实验组之间心输出量及心脏每搏量亦有显著差异。结论 :干细胞移植可明显改善缺血心肌的心脏功能。干细胞移植有望为心肌损伤提供一条新的治疗途径。Objective:Implanting autologous skeletal muscle stem cell- satellite cells into ischemic myocardium to reinforce its structure and function. In this study a reproducible method for the creation of a myocardial lesion wasdeveloped. The functional benefit of cell implantation was evaluated by Necon-3 Noninvasive Cardiac Output Continuous Monitor(USA).Methods:Satellite cells isolated from the skeletal muscle ofadult canines are labeled with 4', 6-dimidino-2-phenylindone(DAPI),which bands to DNA and to the protein tubulin to form a fluorescentcomplex. A ligation of left anterodecendant artery (LAD) was carried out on a canine model to induce a well-delineated transmural lesion.Thereafter, autologous skeletal muscle cells or culture media (control) were infused into ventricular myocardium of isogenic canine through left anterodecendant artery(LAD). Two, four, and eight weeks after cell implantation, 2, 4, 8 weeks later after satellite cell implantation, Cardiac output (CO) and stroke volume (SV) were evaluated by Necon-3 Noninvasive Cardiac Output Continuous Monitor(USA).Results:Necon-3 Noninvasive Cardiac Output Continuous Monitorstudies showed a significant improvement in global and regional left ventricular function in cell-treated canines. There was significant difference in cardiac output (CO) and stroke volume (SV) between the transplantation and the control groups: 2 weeks later, 4 weeks later and 8 weeks later.Conclusions:Satellite cells implanted in the myocardial lesion survived for a 2-month period and were associated with a significant functional improvement of both local and global contraction.
