摘  要:目的 :评价葡萄糖苯脎分光光度法 (简称GPS法 )测定糖化血清蛋白的准确性和特异性。方法 :用GPS法和常用的四氮唑蓝比色法 (简称NBT法 )同时测定临床评价血清中的糖化血清蛋白 ,比较 2种测定方法的重复性、线性、抗干扰能力和临床样本符合情况。结果 :GPS法和NBT法的线性回归系数分别为 0 .96 9和0 .977。 2种方法不同含量样本的批内变异系数均 <5 %。黄疸、脂血及溶血对 2种方法的干扰差异无显著性(P >0 .0 5 ) ,高糖血清 (血糖 >6 .5mmol/L)对GPS法有 5 %~ 10 %的正干扰 ,对NBT法的干扰不明显。 2种方法检测临床样本的相关系数为 0 .895 (P <0 .0 5 )。结论 :GPS法具有良好的线性和重复性 ,同时与NBT法有良好的相关性。Objective:To evaluate accuracy and specificity of glucose phenylosazone spectrophotometric(GPS) method for measuring the glycosylated serum protein(GSP).Methods:Using the GPS method and the nitroblue tetrazalium(NBT) colorimetric method, GSP in the test serum was detected simultaneously. Two methods were compared by observing their agreement for test results, anti-interference ability, intra-assay variability, and linearity.Results:The regression coefficient of linearity of the GPS method and the NBT method was 0.969 and 0.977,respectively. The inter-assay coefficient of variation of different samples detected by two methods was below 5%. The interference of jaundice, lipohemia,and hemolysis serum for the GPS method corresponded with that for the NBT method.The hyperglycemia serum (blood glucose >6.5 mmol/L) was found to be 5%~10% positive interference for the GPS method, interference-free for the NBT method.A correlation co-efficient between the results of GSP as detected by the two methods was 0.895 ( P <0.05).Conclusions:The GPS method is reproducible and highly sensitive with a good correlation with the NBT method.
