摘 要:[目的]观察长期注射胰岛素(INS)及喂食福辛普利对自发性高血压大鼠(SHR)血压、血浆INS浓度、血浆血管紧张素Ⅱ(AngⅡ)浓度、心肌AngⅡ含量及心肌血管紧张素Ⅱ受体1(AT1-R)基因转录的影响。[方法]32只10周龄SHR随机分为4组,1A组:注射INS(5U·kg^(-1)·d^(-1)),服福辛普利(5mg·kg^(-1)·d^(-1));2A组:注射蒸馏水(0.05mL·kg^(-1)·d^(-1)),服福辛普利(5mg·kg^(-1)·d^(-1));1B组:注射INS(5U·kg^(-1)·d^(-1)),服安慰剂(5mg·kg^(-1)·d^(-1));2B组:注射蒸馏水(0.05mL·kg^(-1)·d^(-1)),服安慰剂(5mg·kg^(-1)·d^(-1))。实验前及实验中每2周测大鼠尾动脉血压,9周后抽血并处死动物取材,测量血浆INS、血浆AngⅡ、心肌AngⅡ含量及心肌AT1-RmR-NA水平。[结果]注射INS的1A、1B两组血浆INS较注射蒸馏水的2A、2B两组高(P<0.01);1A与1B间及2A与2B组间SHR的血压、心肌AngⅡ含量、心肌AT1-R mRNA比较无显著性差异(P>0.05);用福辛普利的1A、2A两组SHR的血压、心肌AngⅡ含量及心肌AT1-R mRNA水平均较服安慰剂的1B、2B两组低(P<0.05);1A组与2A组血浆AngⅡ浓度比较无显著差异(P>0.05);1B组血浆AngⅡ浓度显著高于2B组(P<0.05)。[结论]长期注射INS对SHR血压、心肌AngⅡ含量及心肌AT1-R基因转录水平均无显著影响;[Objective] To observe the effects of long-term injection of insulin (INS) and feeding of fosinopril on blood pressure, concentration of plasma INS, plasma angiotensin(AngII), myocardium Ang II and angiotensin II type 1 receptor( AT1-R) gene transcription of myocardium in spontaneously hypertensive rats(SHR) . [Methods] 32 ten-week-old SHR were randomly divided into 4 groups, 1A group; injecting INS (5 U · kg-1 · d-1)andfeedingfosinopril(5 mg · kg-1 · d-1) ;2A group: injecting distilled water (0. 05 mL · kg-1 · d-1)and feeding fosinopril(5 mg · kg-1 · d-1) ;1B group; injecting INS(5 U · kg-1 · d-1) and feeding placebo ( 5 mg · kg-1 · d-1) ;2B group; injecting distilled wa-ter(0. 05 mL· kg-1 · d-1) and feeding placebo(5 mg · kg-1· d-1) . Before and after the study BP was measured by tail-caff method every 2 weeks. After 9 weeks the blood and total mRNA were extracted from myocardium of SHR. The plasma INS, plasma and myocardium Ang II, myocardium AT1-R mRNA were measured. [Results] Plasma INS levels of the INS injection groups were significantly high- er than that of the distilled water injection groups (P <0. 01). The BP, Ang II and AT1 -R mRNA levels of myocardium in the INS injection groups were not significant different from that of the distilled water injection groups respectively ( P > 0. 05). The BP and Ang II and AT1 -R mRNA levels in the fosino-pril feeding groups were significantly lower than that of the placebo group respectively (P < 0.05) . The level of plasma AngII in 1B group was significantly higher than that in 2B group( P < 0. 05) . [Conclusion] Long-term injection of INS has no significant effects on BP, myocardium Ang II and gene transcription of myocardium AT1-R in SHR. The BP is decreased and myocardium Ang II and gene transcription of myocardium AT1-R in SHR are dowm-regulated by fosinopril.
