摘  要:目的 建立一个简便的研究延迟性排斥反应的猪—猴异种心脏移植模型。方法 通过流式细胞仪法监测猕猴体内天然抗体水平在出生后 0 .5~ 3月龄的变化 ,选择新生梅山猪和 2 .5月龄中国猕猴分别作为供、受体 ,建立非协调性猪—猴异种移植模型。观察移植心存活时间 ,排斥后取移植心进行电镜、病理及免疫组化检查。结果 IgM类天然抗体水平出生后缓慢上升 ,3月龄时达到 31%左右 ,而IgG类天然抗体水平始终处于低水平。移植心脏平均存活时间 36h ,电镜、病理及免疫组化显示为延迟性排斥反应。结论 选择新生梅山猪和 2 .5月龄中国猕猴建立非协调性猪—猴异种移植模型 ,可方便克服急性排斥反应 ,从而有助于延迟性排斥反应的研究。Objective To establish a simple pig-to-monkey xenograft model to study delayed xenograft rejection (DXR).Methods The change of natural antibody (IgM and IgG) between 0.5-3 months old in the monkey was detected by the flow cytometry.The newborn Meishan pig and 2.5 months old rhesus monkey were served as donors and recipients respectively to establish discordant pig-to-monkey xenograft model.Mean survival time (MST) was measured after xenotransplantation.The rejected xenografts were analysed by electronic microscope,pathology and immunobiolochemistry.Results The level of natural antibody IgM ascended slowly after birth,and get to 31% at 3 months old,but the level of natural antibody IgG was at low level all the time.The MST of xenografts was 36 hours,and we found it was DXR by electronic microscope,patology and immunobiolochemistry.Conclusions The newborn Meishan pig and 2.5 months old rhesus monkey can be used to establish a pig-to-monkey discordant xenograft model to overcome hyperacute rejection(HAR) easily and study DXR.
