免疫调节剂SCA诱导的抗血吸虫性肝病变及其机制的研究

摘  要:目的 探讨感染日本血吸虫的BALB/c小鼠诱导的抗肝内血吸虫卵肉芽肿病变效应及其分子机制。方法 取鼠肝连续切片 ,HE染色 ,光镜下测量肉芽肿大小 (平均面积和体积 ) ;用流式细胞仪测定鼠脾T淋巴细胞亚群CD4 + 和CD8+ 百分数及二者的比值 ;用ELISA检测小鼠血清特异性抗体IgG及其亚类IgG1和IgG3 水平。结果 服用SCA(SCA - 1,SCA - 2 )实验小鼠肝内肉芽肿病变均非常显著地小于感染对照组 (均P <0 .0 1) ;实验鼠肝内虫卵肉芽肿的平均面积和体积 ,与对照组比较 ,减小率达 4 0 .71%~ 5 3.0 8% ;实验组CD4 + /CD8+ 值 (2 .72~ 3.6 7) ,显著地高于对照组 (1.89) (均P <0 .0 5 ) ;用药组的特异性抗体IgG及其亚类IgG1和IgG3 水平 ,均显者地高于对照组 (均P <0 .0 1或 0 .0 5 )。结论 中药合剂SCA是一种新型、高效无毒的免疫调节剂 。Objective To study the immunoregultory effects and mechanism of anti-Schistosoma japonicum hepatic egg granuloma formation induced by Immunomodulator SCA. Methods The mean area and volume of schistosome egg granuloma (SEG)were measured by morphometric examination of liver tissue in mice.The percentage of splenic lymphocyte phenotypes of mice was detected by flowcytometer The serum specific antibody IgG and its subclass IgG 1 and IgG 3 levels were determined by ELISA. Results The mean sizes of SEG in the experimental groups were more significantly decreased than that in the control group ( P <0.01).The reduction rate of SEG had no marked difference in the orally-administered orally groups ( P >0.05).The mean sizes of SEG were decreased from 40.71% to 53.08% in the two experimental groups.The ratio of CD4 +/CD8 + in the experimental groups (2.72 and 3.67) was obviously higher than that in the control group(1.89),The lgG,IgG 1 and IgG 3 levels of the experimental groups were markedly higher than that in the control group, P <0.01 or P <0.05. Conclusion It is concluded that SCA is a new,effective and non-toxic immuno-modulator,and its molecular mechanism is closely associated with the enhancement of cellular and humoral immune responses.
