摘 要:目的 观察纳络酮对兔缺血再灌注心肌细胞线粒体呼吸耗氧量和游离脂肪酸含量的影响。方法 建立兔心肌缺血再灌注模型、分离线粒体 ,用氧电极法检测线粒体氧耗量和用气相色谱分析法测定游离脂肪酸含量。结果 单纯缺血组生理盐水和纳络酮治疗心肌线粒体呼吸氧耗量与对照组无明显差别 (P >0 .0 5 ) ;缺血再灌注组生理盐水和纳络酮治疗心肌线粒体呼吸耗氧量明显低于单纯缺血组和对照组 (P <0 .0 5 ,P <0 .0 1 ) ,但纳络酮治疗组明显高于生理盐水治疗组 (P <0 .0 1 ) ;单纯缺血组、缺血再灌注组生理盐水和纳络酮治疗心肌线粒体FFA含量均明显高于对照组 (P <0 .0 5 ,P <0 .0 1 ) ;再灌注时纳络酮治疗心肌线粒体FFA含量明显低于生理盐水治疗心肌 (P <0 .0 5 )。结论 纳络酮能够提高缺血再灌注后心肌细胞线粒体氧耗量、降低游离脂肪酸含量 。Objective To investigate the influences of naloxone on O 2 consumption and FFA content in mitochondria of rabbit's myocardium following ischemia and reperfusion. Methods The model of myocardial ischemia and reperfusion was set up and mitochondria of myocardium was isolated,the changes of the O 2 consumption of mitochondria was detected by oxygen electrode method and the content of FFA was extracted and assayed by the method of gas chromatography. Results The O 2 consumptions of mitochondria in ischemic groups treated with saline and naloxone had no significant difference comparing with control groups( P <0.05), the O 2 consumptions of mitochondria in ischemia and reperfusion groups treated with saline and naloxone were significantly decreased comparing with control groups and ischemic groups( P <0.05, P <0.01),but the O 2 consumptions of mitochondria in groups treated with naloxone was higher than that in group treated with saline following ischemia and reperfusion; the contents of FFA in mitochondria in groups treated with saline and naloxone in ischemia or following ischemia and reperfusion were higher than that in control groups( P <0.05, P <0.01),but it in group treated with naloxone following ischemia and reperfusion was lower than that in group treated with saline( P <0.05). Conclusions Naloxone might be able to increase O 2 consumption and reduce FFA content in myocardial mitochondria,it suggested that naloxone have a protective effect on myocardial mitochondria.
