摘  要:选用猪 新生猴腹腔心脏异位模型 ,通过异种胸腺修饰加60 Coγ照射途径 ,来探讨此途径对异种器官移植中T细胞的作用及诱导异种T细胞中枢性耐受的可能性 ,并进一步研究外周血IL 2水平与排斥反应的关系。结果发现 :MLR检测在照射 +胸腺注射组接受脾细胞胸腺内注射后第 3周 ,其刺激效应较空白组、照射组下降明显 (P <0 0 1 )。照射 +胸注组存活期较空白组明显延长 (P <0 0 1 ) ,照射 +胸注组存活期较胸注组和照射组延长 (P <0 0 5 )。IL 2和IL 1 0检测的结果表明 ,各组在排斥时均表现为IL 2水平较移植前显著升高 (P <0 0 0 1 ) ,胸注 +照射组和胸注组 ,受体体内IL 2水平 ,在移植后早期与移植前比较无显著性差异 (P >0 0 5 ) ,与同时期单纯照射和空白组术后比较有显著差异 (P <0 0 1 )。结果表明 :异种胸腺注射可诱导供体特异性的T细胞功能抑制 ,异种胸腺注射联合照射可有效地延长供心存活时间。IL 2水平与排斥反应的发生关系密切 。To investigate the validation of pig to monkey as an experimental animal model for the study on the induction of immune tolerance to cardiac xenografts by intrathymic inoculation with xenogeneic spleen cells and whole body γ ray irradiation,and to determine the correlation between the rejection of donor heart and the level of IL 2,the present investigation was done in our laboratory The experimental results of this study showed,the survival times of donor heart in the irradiation and intrathymic inoculation groups (91 1±22 18) hours were significantly longer than those of blank group (36 6±5 8) hours In MLR,when the recipient splenocytes responded to the donor stimulator,there was significant reduction in the irradiation and intrathymic inoculation group than in the blank group as well as the whole body irradiation group The levels of IL 2 of recipients of all groups were much higher in the rejection of xenografts than those before transplantation.These results suggest that pretreatment with intrathymic inoculation and whole body irradiation could induce T cell immune suppression and the changes of serum IL 2 levels are closely correlated with the development of xenograft rejection It could be used as an important index to inspect to the late rejection reactions
