摘  要:目的 研究视神经损伤后睫状神经营养因子 (ciliaryneurotrophicfactor,CNTF)及其受体 (CNTFRα)蛋白在大鼠视网膜中的表达变化。 方法 采用钳夹法制作大鼠视神经损伤模型 ,分别于伤后 1,3,7,14 ,2 8d取眼球 ,以正常大鼠视网膜为对照 ,提取视网膜蛋白 ,采用Westernblotting法检测视网膜中CNTF和CNTFRα蛋白的表达变化。 结果 在正常视网膜中存在CNTF和CNTFRα蛋白的表达。视神经损伤后 1,3,7,14及 2 8dCNTF和CNTFRα蛋白表达均增高 ,伤后 7d达到最高 ,14d下降。CNTF蛋白表达量至 2 8d仍显著高于正常组 (P <0 .0 5 ) ,而CNTFRα蛋白表达量至 2 8d虽增高 ,但与正常对照组比较 ,差异无显著性意义 (P >0 .0 5 )。 结论 视神经损伤后视网膜中CNTF和CNTFRα蛋白表达增加 ,可能是视网膜神经元自我保护的一种反应。Objective To study the expression changes of ciliary neurotrophic factor (CNTF) and ciliary neurotrophic factor receptor alpha (CNTFRα) following optic nerve injury in rats. Methods The optic nerve injury model of rats was induced by crushing 5 mm optic nerve retrobulbarly. The eyeballs were removed at day 1, 3, 7, 14 and 28 respectively after optic nerve injury, using normal rats as control. Western blotting method was used to detect the protein levels of CNTF and CNTFRα in the retina. Results There existed protein expression of CNTF and CNTFRα in the normal retina. The protein expressions of CNTF and CNTFRα were increased significantly at day 1, 3, 7, 14 and 28 after injury, reached peak at day 7 and began to decrease at day 14.The protein expression of CNTF was still significantly higher than that in the control group at day 28 (P<0.05). Nevertheless, the protein expression of CNTFRα was increased at day 28 with no significant difference compared with the control group (P> 0.05). Conclusions The protein expression increase of CNTF and CNTFRα in the retina after optic nerve injury may be a self-protection response of the retinal neurons.
