运用血清药理学方法研究红花对豚鼠离体心脏的作用

摘　　要：目的观察对家兔灌胃给予红花提取物后 ,其含药血清对豚鼠离体心脏的作用。方法家兔每日 2次灌胃给予红花提取物 5、1 0、2 0 g·kg-1,连续 3d。末次给药后 60min取血 ,分离血清。将含药血清加到豚鼠离体心脏系统 ,观察红花对离体心脏冠脉流量 ,心搏幅度及心率的影响。同时 ,采用高效毛细管电泳方法测定含药血清指纹图谱。结果灌胃给予红花提取物 5、1 0、2 0 g·kg-1后 ,含药血清均能显著增加豚鼠离体心脏冠脉流量 ,增强心搏幅度及减慢心率 (P <0 0 5或P <0 0 1 )。在灌胃给予红花提取物后 60min所采血清的指纹谱中出现一些新的峰。结论红花能增加冠脉流量 。Objective To observe the effect of Flos Carthami on the guinea pigs′ hearts in vitro.Method The experiment was accomplished by serum pharmacology.Different ig dosages (5,10, 20 g·kg -1) were administered to rabbits twice per day on three consecutive days.The blood was taken at 60 min after the last administration and the serum was separated.The separated serum was added to the guinea pigs′ hearts in vitro.The influence of the separated serum on the coronary artery flow, heart stroke amplitude and heart rate was observed.The fingerprint of the serum was determined by the high performance capillary electrophoresis.Results The separated serum from administered rabbits was observed to increase the coronary artery flow, reinforce the heart stroke amplitude and decrease the heart rate of the guinea pigs′ hearts significantly in vitro(P<0 05 or P<0 01).Some peaks appeared newly in the fingerprint of the serum from the administered rabbits.Conclusion Flos Carthami has the effect on increasing the coronary artery flow, enhancing the heart stroke amplitude and decreasing the heart rate.
