摘 要:目的探讨鲑鱼鱼白DNA(salmonmiltDNA,SMD)对小鼠胸腺增龄性萎缩的作用及作用机制。方法10月龄雌性BALB/c小鼠按体重随机分成缓冲液对照组(controlgroup,C组)、低剂量组(lowdosagegroup,L组)和高剂量组(highdosagegroup,H组),每组26只。在标准饲料基础上分别每日灌胃0.1mol/L的柠檬酸钠缓冲液和166.67mg/(kg·d),333.33mg/(kg·d)的SMD。5周后,无菌取胸腺,测量胸腺脏器指数;每个胸腺单独蜡块包埋,切片用Image-proPlus专业图像分析系统(4.0版)进行细胞计数和皮质厚度测量并经SAS(6.12版)统计软件分析数据。应用基因芯片技术在C组和H组胸腺组织中筛选表达差异的基因片段、RT-PCR鉴定部分片段。结果SMD对小鼠的体重、胸腺重量和胸腺指数均无影响(各指标F<3.0,P>0.05);高剂量组小鼠的胸腺皮、髓质细胞数量与对照组相比均显著增多犤皮质D(H,C)=9.46,P<0.01;髓质t(H,C)=2.53,P<0.05犦;低剂量组的胸腺皮、髓质细胞数量与对照组相比差异均不显著犤皮质D(L,C)=3.65,P>0.05;髓质t(L,C)=0.8,P>0.05犦;高、低剂量组的胸腺皮质平均厚度均显著高于对照组犤t(H,C)=4.01,P<0.01;t(L,C)=2.80,P<0.05犦;经基因芯片技术初筛出112条差异表达的基因片段;Genebank登录号为Aw209102,U23789,X80232的3条上调表达的基因片段。Aim To investigate the effect and the m ec hanism of salmon milt DNA(SMD)on age-related involutions in mouse thymus.Met hods Female BALB /c of 10months were divid ed randomly into three groups acc ording to their weights:high dosage group 333.33mg /(kg·d),low dosag e group 166.67mg /(kg·d)and control group 0mg /(kg·d).After five weeks,with Image-Pro Plu s(version.4.0)software,the thymus indexes and the thymoctytes in the th ymus section were measured,as well as the thymus cor tex thickness.All t he data were analyzed by SAS statistic software.Microarray technique was applied to screen the gene fragments,which were differently expr essed between the high dosage group and the control group,together with RT-PCR to further confirm some of them.Results No significant differences of the vari ables including bodyweight,thymus weight and thymus ind exes among the three gr oups were found(F<3.0and P>0.05,respectively).The thymocytes quanti -ties of thymus cortex and medulla in t he high dosage group were significan tl y higher than those of the control grou pcortex D (H,C ) =9.46,P<0.01;medulla t (H,C) =2.53,P<0.05.The thymus cortex thicknesses of both SMD supplem ent groups were signific antly higher than that of the control groupcortex D (L,C ) =3.65,P>0.05;medulla t (L,C ) =0.8,P>0.05.112differently expressed gene fragmen ts were is olated.Furthermore,we fo und the fragments with the logged number of U2378 9,X80232and Aw209102were highly expressed in the high dosage group when RT-PCR technique was used.Conclusion SMD may reverse the age-related inv o lutions in mouse thymus via up-regulation the e xpression of proliferation re lated genes and development and differentiatio n related genes simultaneously.
