摘  要:目的:探讨电针对坐骨神经损伤大鼠的脊髓中神经生长相关蛋白-4(GAP-43)mRNA表达的影响。方法:60只Wistar大鼠制成坐骨神经损伤模型,经电针穴位刺激治疗后用原位杂交技术观察坐骨神经相应脊髓节段GAP-43mRNA的表达。结果:电针组第1天阳性细胞数增多(23.5±4.1)个/mm2,3d达高峰(46.8±6.8)个/mm2,14d后明显减少(9.7±3.6)个/mm2,28d后基本未见阳性细胞存在(1.0±1.2)个/mm2;模型组第1天(16.3±2.9)个/mm2至第7天(20.1±2.5)个/mm2持续阳性细胞数增多,1d略减少(19.1±3.2)个/mm2,28d后仍可见阳性细胞存在(8.5±2.0)个/mm2。与电针组比较差异有显著性意义(t=2.36~4.25,P0.05)。结论:穴位电针能增强并缩短GAP-43mRNA表达时间,有利于突触重建。AIM:To explore th e effects of electric acupuncture on the expression of GAP-43mRNA in spinal m otoneurous of sciatic nerves in rats.METHODS:Sixty male adult Wistar rats were enr olled to establish injured sciatic nerve models and equally div ided into a cupuncture group and model group.The rats were treated by electric acupuncture .The expressions of GAP-43mRNA in spinal cord were exami ned by in situ hybr idization.RESULTS:In the acupuncture group,the number of GAP-43mRNA pos-i tive cells increased[(23.5±4.1)/mm 2 ]at the first day,and up to the peak(46.8±6.8)/mm 2 at the 14th day,but decreased[(9.7±3.6)/mm 2 ]after 14d,and GAP-43mRNA positive c ells[(1.0±1.2)/mm 2 ]were hardly found after 28d.In the model g roup,the number of GAP-4 3mRNA positive cells increased from(16.3±2.9)/mm 2 at the 1st day to(20.1±2.5)/mm 2 at the 7th day continuously till the 28th day[(8.5±2.0)/mm 2 ],and GAP-43mRNA positive cells were still found after 28d.A signifi cant difference was presen t between the two groups(t=2.36-4.25,P< 0.05).
