脑栓康的抗脑缺血作用及其机制研究

摘　　要：目的:评价脑栓康对脑缺血的防治作用,并探讨其作用的可能环节。方法:采用大鼠双侧颈动脉结扎造成脑缺血模型,测定脑含水量、K+、Na+及丙二醛(MDA)含量,并进行组织形态学检查;测定正常和血瘀大鼠血小板聚集活性、正常大鼠脑血流量及血瘀大鼠血液流变学指标。结果:脑栓康150mg/kg可明显降低脑缺血大鼠的脑含水量和脑MDA含量(P<0.05);脑栓康75mg/kg可减轻脑缺血大鼠脑组织的Na+蓄积和K+流失(P<0.05),可使缺血脑组织海马旁回神经元变性明显改善;脑栓康对大鼠血小板聚集有明显的抑制作用,一次给药150mg/kg、75mg/kg对ADP诱导的正常大鼠血小板的聚集抑制率分别为85.6%和72.8%,脑栓康300,150和75mg/kg连续给药5d,对ADP诱导的血瘀大鼠血小板的聚集抑制率分别为62.6%、57.3%和32.8%;脑栓康300mg/kg1次给药可使正常大鼠的脑血流量增加9.6%,也可使血瘀大鼠血浆纤维蛋白原含量由(7.20±1.49)g/L降低至(6.03±0.88)g/L。结论:脑栓康有明显的抗脑缺血作用,其作用环节可能有:抑制血小板聚集、降低血液纤维蛋白原含量、增加脑血流量和抗氧化减少细胞损伤等。AIM:To evaluate the effe ct of Naoshuankang in prevention and cure of cerebral ischemia and to study its me chanism.METHODS:Bilateral carotid ligation models o f rats were established t o test the contents of K + ,Na + ,H 2 O and MDA in brain,and examine brain t issue morphology under microscope.T he hemorrheologic indexes,platelet aggre-gation and cerebral blood flow were m easured in the model and normal rats.RESULTS:Naoshuankang(75mg /kg)signif icantly prevented K + loss and Na + accumulation in acute cerebral isch emic rats(P<0.05),and ame-liora ted the neuronal degeneration of Hippocampus of ischemic brain tis sue.The brai n water and MDA contents were decreased with Naoshuankang of150mg /kg(P<0 .05).Administraction of 150mg /kg and 75mg /kg Naoshuankang obviousl y inhibited t he platelet aggregation of the norma l rats induced by ADP with th e inhibitive ra te of 85.6%and 72.8%,respective-ly.Treated with 3 00mg /kg,150mg /kg and 75mg /kg Naoshuankang for5d the platelet aggr egation of the mo del rats were inhibited with the in-hibitive rate of 62.6 %,57.3%and 32.8%,respectively.Administration o f300mg /kg Naos huankang increased th e brain blood flow of the normal rats by 9.6%and decre ased the plasma fibr inogen content of the model rats from(7.20±1.49) g /L to(6.03±0.88)g /L.
