治疗用武汉(Wu)抗人T淋巴细胞单克隆抗体与人体组织反应性的体外研究  

The Reactivities of Wuhan Anti-human T Lymphocyte McAbs for Therapeutical Use with Normal Human Tissues in Vitro

摘　　要：应用免疫组化法及免疫细胞荧光法体外检测了WuT_3(抗CD_3)、WuT_4(抗CD_4)、WuT_3(抗CD_3)、WuT_9(抗CD_(71)或TfR)及WuTac(抗CD_(25)或IL-2R)等5种小鼠抗人T淋巴细胞单克隆抗体与25种正常人体组织的反应性。结果显示,上述单抗均与淋巴组织中各自靶细胞呈特异性阳性反应。此外,睾丸中散在分布个别WuT_3阳性淋巴细胞,WuT_9与26％的骨髓细胞及小肠粘膜肠腺底部细胞是阳性反应。本文尚探讨了上述单抗在临床治疗器官移植排斥反应等中的应用价值。By the immunohistological enzyme staining and immunocytological fluorescent staining, the reactivities of WuT3(anti-CD3), WuT4(anti-CD4), WuT8(anti-CD8), WuT9(anti-CD71 or TfR) and WuTac(anti-CD25 or IL-2R) McAbs with 25 normal human tissues in vitro were detected. The results showed that WuT3, WuT4 and WuT8 reacted respectively with their target cells in lymphoid tissues with high specificity. Besides, there were a few WuTs positive lymphocytes scattered in testis. When stained with WuT9, 26% of bone marrow cells, a few scattered cortical thymocytes and the bottom cells of intestinal mucosa gland were positive. WuTac reacted with a few mo-dullary thymocytes. All of the 5 McAbs for therapeutical use showed negative reactions with other non-lymph tissues and cells.
