抗碱性磷酸酶单克隆抗体杂交瘤细胞系的建立及其APAAP的制备与初步鉴定

摘　　要：本文报道用碱性磷酸酶Sigma Ⅰ型粗酶免疫Balb/c小鼠,采用酶联免疫吸附反应筛选细胞融合后杂交瘤培养上清,最后获7株稳定分泌抗碱性磷酸酶单抗的杂交瘤细胞株,对其中6株进行了特性鉴定。取其中1株AAP1/323腹水制成APAAP试剂盒,应用于ELISA和免疫组化染色表明:该试剂盒灵敏度高,本底低,无内源性酶干扰现象,对比好,背景清晰,结果容易判断,能满足临床检测的要求。Establishment of mouse hybridoma secreting monoclonal antibodies to alkaline phosphatase and preparation and evaluation of APAAP are described in this paper. The clones were screened with reverse indirect enzyme linked immunoadsorp-tion assay and finally seven clones steadily secreting antibodies were obtained. Of them,six were investigated in characterizations. AAP1/323 ascities of them was mixed into APAAP with alkaline phosphatase for applications of ELISA and immuno-histochemistry staining.The results of tests displayed that the APAAP kit is of characterizations of high sensitirity, the lowest background and no interference with endogenous enzymes and other significant advantages including easy evaluation due to high color contrast of the enzymatic reaction, availabilty of non-toxic substrate reactions.
