摘 要:猪的毛色表型虽然与经济性状没有直接相关 ,但它却对经济效益产生重要影响 ,在猪育种实践、商品猪生产等方面都有应用。结合PCR AccⅡ RFLP、PCR BspHⅠ RFLP及PCR SSCP技术 ,分析了 16个全同胞家系和金华猪、嘉兴黑猪、玉山黑猪、乐平花猪、上高两头乌猪及嵊县花猪等 6个地方猪种随机采样个体的黑素皮质素受体 1(MC1R)基因型。结果显示 ,地方猪种在MC1R位点携带高频率的显性黑等位基因ED1 ,表明我国地方猪种的黑毛色可能主要由显性黑等位基因ED1 调控。通过对嵊县花猪MC1R位点的分析 ,首次发现PCR SSCP证据的新序列 ,与已知的其他 5个等位基因带型不同。家系个体的分析结果进一步验证了ED1 对EP、e为完全显性 ,EPAlthough coat color in pigs has no direct relatio n with economic traits,it affects economic benefit significantly,coat color select ion are widely used in pig breeding and production.PCR-AccⅡ-RFLP、PCR- BspHⅠ-RFLP and PCR-SSCP were used in combination to analyze genotype at MC1R locus among individuals from 16 full-sib pedigre es and 6 Chinese native breeds including Jinhua,Jiaxing Black,Yushan Black,Lepin g Spotted,Shanggao Spotted and Shengxian Spotted pig.It was found that the Chine se native pig breeds carry a dominant black allele at MC1R at high frequency,this E D1 allele was suggested to be the major allele controlling black coat color in Chinese native pig breed.In addition,the evidence for a new allele was obtained in Shengxian Spotted pigs by PCR-SSCP a nalysis.It was reconfirmed from the result of pedigree analysis that E D1 was dominant over E P and e,while E P was incompletely dominant over e.
