摘  要:目的探讨谷氨酰胺(GLN)二肽胃肠外营养对短肠大鼠小肠上皮细胞产生营养作用的机制。方法18只Wistar大鼠,手术切除其大部分空肠及近端回肠,然后经右颈内静脉插管,建成短肠大鼠胃肠外营养模型;大鼠随机分为3组:常规胃肠外营养组(STD组),3%丙氨酰-谷氨酰胺胃肠外营养组(GLN组)和正常对照组(n=4),连续7d胃肠外营养后,在实验结束前经颈静脉注入5-溴脱氧尿苷(BrdU),采用ABC方法及光镜观察方法。结果GLN组小肠PCNA阳性细胞数多于STD组,差异有显著性(P<0.05)。GLN组小肠BrdU阳性细胞数多于STD组,差异有显著性(P<0.05)。结论提示GLN二肽可促进短肠大鼠小肠上皮细胞的分裂增殖。Objective To study the nutrient mechanism of glutamine dipeptide-enriched parenteral nutrition on rat's residual small bowel following extensive small bowel resection.Methods Using18Wistarrats,the model of parenteral nutrition was established after extensive small bowel resection and intubating via right internal jugular veins.These rats were randomly divided in to three groups:standard group(STD group),3%alanine-glutamine-dipeptide group(GLN group),normal group (n=4).After intravenous infusing enriched parenteral nutrition continuously for7days;then the samples of residual small bowel were taken after intravenous injection of5-bromodeoxyuridine(BrdU)from internal jugular vena,ABC immunohistochemistry assay and light microscopy were performed.Results Proliferating cell nuclear antigen(PCNA)and BrdU-labelled cells were predominantly in crypt epithelium of small bowel in each rat.Compared with that of the STD group,the numbers of the PCNA and BrdU labelled cells per crypt of GLN group increased significantly(P<0.05).Conclusion Glutamine dipeptide enriched parenteral nutrition can improve the residual intestinal epithelium proliferating.
