摘  要:目的 研究人巨细胞病毒 (humancytomegalovirus,HCMV)体外感染对人脐静脉内皮细胞 (HUVEC)表面细胞间粘附分子 1(ICAM 1)mRNA和核转录因子kappaB(NF κB)表达的影响 ,探讨NF κB活化与ICAM 1mRNA表达的关系。方法 用RT PCR方法检测ICAM 1mRNA的表达 ,用凝胶电泳迁移率改变试验 (EMSA)检测NF κB活性。结果 HUVEC感染HCMV后 1h ,NF κB活性增加 (P <0 .0 5 ) ,感染后 2h ,ICAM 1mRNA表达增加 (P <0 .0 5 ) ,二者均于 8h达高峰 ,至 2 4h时仍维持较高表达(P <0 .0 1) ;加入NF κB活性抑制剂四氢吡咯二硫代氨基甲酸酯 (PDTC)后 ,NF κB活性明显减弱 ,同时ICAM 1mRNA的表达也明显降低 (P <0 .0 1)。结论 HCMV感染可激活NF κB ,使ICAM 1mRNA的表达增加 ,从而引导炎症反应至感染部位 ,并有利于病毒感染在细胞间扩散。NF κB参与ICAM 1转录的调控 ,其活性变化影响ICAM 1mRNA的表达。Objective To investigate the effect of human cytomegalovirus (HCMV) infection on the expression of intercellular adhesion molecule-1 (ICAM-1) mRNA and nuclear factor kappa B (NF-κB) in human umbilical vein endothelial cells (HUVEC) so as to explore the relationship between the activation of NF-κB and the expression of ICAM-1 mRNA. Methods HUVEC was cultured and then infected by HCMV. Expression of ICAM-1 mRNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Activity of NF-κB was measured by electrophoretic mobility shift assay (EMSA). Results One hour after infection (h.p.i) by HCMV, the activity of NF-κB increased significantly as compared with mock-infected cells (P<0.05); at 2nd h.p.i, the expression of ICAM-1 mRNA also enhanced (P<0.05); both reached the peak at 8 h.p.i and remained relatively high level at 24th h.p.i (P<0.01). When the specific NF-κB inhibitor pyrrolidine dithiocarbamate (PDTC) was added, the activity of NF-κB dramatically decreased while the expression of ICAM-1 mRNA also decreased significantly (P<0.01). Conclusions The infection with HCMV may activate NF-κB and thereby enhance the expression of ICAM-1 mRNA in HUVEC which target the inflammatory reaction to sites of infection and facilitate the dissemination of HCMV among HUVEC. NF-κB may play a role in the regulation of the transcription of ICAM-1 and the change of its activity may influence the expression of ICAM-1 mRNA.
