摘 要:目的 观察褪黑素抗大鼠慢性肝损伤作用。方法 用四氯化碳 (CCl4)制造大鼠慢性肝损伤模型 ,同时予褪黑素腹腔注射 ;在造模的第 1、2、3月末处死大鼠 ,分别测定大鼠体重、肝脏湿重和血清丙氨酸转氨酶 (ALT)、天冬氨酸转氨酶 (AST)及白蛋白和总蛋白水平 ,并作肝脏病理学检查。结果 褪黑素可以显著降低CCl4诱导大鼠慢性肝损伤早期的ALT、AST ,在大鼠慢性肝损伤的中期 ,MT可提高血清白蛋白水平和白球比值 ,改善肝脏病理损伤。结论 褪黑素在CCl4诱导的大鼠慢性肝损伤早期可以减轻肝脏炎症 ,而在中晚期 。Objective To observe effects of melatonin against chronic hepatic injury in rats continuously. Methods Rat chronic hepatic injury model was induced by carbon tetrachloride (CCl 4), and treated with melatonin ip at the dose of 10 mg/kg body weight. The rats were killed at the end of the 1st, 2nd, 3rd month and the rats body weight; wet liver weight and alanine transaminase (ALT), asparagic acid transaminase (AST), albumin, total protein in serum were determined. Histological examination of rats liver was also performed. Results On the early phase of rat chronic hepatic injury melatonin decreased ALT, AST,and increased albumin, total protein in serum significantly on the middle phase, and ameliorated liver pathological changes in rats. Conclusion Melatonin ameliorates rat chronic hepatic injury during the early phase of CCl 4 induced rat chronic hepatic injury; melatonin relieves liver inflammation during the middle or later phases; melatonin mainly playes the role of inhibiting liver fibrosis.
