猪红细胞CD_(58)分子的疫苗佐剂效应研究

摘  要:用从木瓜酶修饰的猪红细胞提取的CD58分子协同NDⅣ疫苗 ,免疫 8日龄雏鸡 (分为试验A组、对照B组 ) ;用从正常猪红细胞提取的CD58分子协同NDⅣ疫苗 ,免疫 18日龄雏鸡 (分为试验C、对照D组 )。以 β_微量法测定ND抗体效价 ,以α_醋酸萘酯酶染色法检测鸡外周血T细胞的变化 ,并分别用F48E9强毒株在免疫后 2 4d攻击A组和B组鸡、39d攻击C组和D组鸡。结果表明 ,免疫前 ,8日龄雏鸡的ND抗体效价为 2 5.8,18日龄雏鸡的ND抗体均为阴性 ;免疫后 ,与对照D组比较 ,试验C组的ND抗体提前 4d达到 2 4.8,并且试验C组ND抗体效价显著高于对照组 (P <0 .0 5或P <0 .0 1) ,保护期延长 ;试验A组雏鸡体内ND抗体变化和对照B组相似 ,但试验A组雏鸡外周血T细胞数量显著高于对照B组 (P <0 .0 1或P <0 .0 5 )。攻毒试验结果显示 ,试验A组鸡死亡率为 33%、对照B组鸡死亡率为 6 3% ;试验C组鸡死亡率为 17% ,对照D组鸡死亡率为 5 3%。本研究结果表明猪红细胞CD58分子具有较好的疫苗佐剂效应。Newcastle disease(ND)Ⅳ strain vaccine mixed with CD_(58) from pig reyghrocytes modified by papain or without that of CD_(58) were injected to 8 days chicks (test group A,control group B);ND Ⅳ strain vaccine mixed with CD_(58) from normal pig erythrocytes or not were injected to 18 days chicks(test group C,control group D).The ND antibody titer was measured with β microdetermination method and T cells in peripheral blood were detected with acid α-nathpyl acetate esterase(ANAE).24 d after inoculation,group A and group B,and 39 d after inoculation,group C and group D were all attacked with F_(48)E_9 NDV strain.The results show,ND antibody titers of 8 d chicks were 2^(5.8) and that of 18 d chicks were all negative before inoculation;After inoculation,ND antibody titers of test group C chicks reached 2^(4.8) 4 d early and were remarable higher (P<0.01,or P<0.05)than that of control group D,the protective period in group C was extended.The number of T cells in peripheral blood of test group A is higher than that of control group B,although the change of ND antibody titers of test group A chicks was similar to that of control group B.F_(48)E_9 NDV strain attack test results show that the chicks death rate of group A was 33 %,group B was 63 %,group C was 17 %,and group D was 53 %.These results indicate that CD_(58) molecule from pig erythrocytes has a good vaccine adjuvant effect.
