摘 要:目的 :探索脂肪组织提取物 (PLA)细胞分化为神经元样细胞的可能性 ,为PLA细胞分化潜能的研究及其在神经科学领域的应用提供依据。方法 :以大鼠的PLA细胞为实验对象 ,利用 β 巯基乙醇 (β BME)、丁酸酯羟基茴香醚 (BHA)等作为分化诱导因子 ,对各阶段细胞进行形态学和免疫组化鉴定。结果 :诱导培养 2h后 ,大多数存活的细胞转变为类似于神经元的形态 ,分化培养基诱导 5h后 ,从简单的双极细胞到复杂的多极细胞均有出现 ,并可见部分神经元样细胞拉成网状 ,大多数存活细胞表现为NSE染色阳性。结论 :PLA细胞在体外培养条件下 ,可以向神经元样细胞分化。提示PLA细胞可能作为一种新的成体多能干细胞在神经科学领域加以利用。Objective: To investigate the feasibility of inducing the differentiation of processed lipoaspirate(PLA) cells into neuron like cells in vitro, to offer further research on the reference for the application of PLA cells in the field of neuroscience. Methods: PLA cells were obtained form rats. Butylated hydronyanisole(BHA) and β Mercaptoethanol (β BME) were used as differentiation factors. Morphology and Immunohistochemistry observation of Cells were employed to identify the cell at different culture stage. Results: After 2 h under differentiation condition, the PLA cells began to appeared as neuron like cells. After 5 h, Neuron like cells from simple 2 to complex multi polar cells all appeared and most of them were NSE positive stained. Conclusion: Under differentiation culture condition, PLA cells can differentiate into neuron like cells in vitro, which indicate that as a new adult pluro, PLA can be used in the neuroscience field.
