摘  要:目的 :观察肾上腺髓质素原来源的两个不同肽段Adrenomedullin (ADM )和 proadrenomedullinN -ter minal 2 0 peptide (PAMP)单独或联合作用对血管张力的影响。 方法 :在孵育的大鼠离体胸主动脉环上预先加入苯肾上腺素使之收缩达最大张力后 ,单独或联合加入不同浓度的ADM和PAMP ,用张力换能器和多导生理记录仪 ,记录其对离体血管环张力作用曲线。结果 :10 - 9~ 10 - 7mol·L- 1的ADM浓度依赖地抑制苯肾上腺素诱导的血管收缩 ;相同浓度的PAMP对苯肾上腺素诱导的血管收缩或基础状态下的血管张力无显著影响。不同浓度的PAMP与ADM联合应用对苯肾上腺素诱导的血管张力反而有增强作用。血管去除内皮或用L -N -硝基精氨酸预先孵育后 ,ADM的舒血管效应及PAMP对其的拮抗作用均消失。结论 :ADM可诱导血管舒张 ,PAMP可拮抗ADM的舒血管效应 。Objective To observe the effects of d ifferent peptides derived from proadrenomedullin, ADM and proadrenomedullin N-te rminal 20 peptide (PAMP), individual or in combination, on vascular ring tone. Methods Isolate rings from rats aorta with or without endo thelium vascular tone were recorded by the precontracted with phenylephrine and put into the ADM and PAMP, individual or in combination. Results ADM at concentrations of 10 -9 ~10 -7 mol·L -1 induced co n centration dependent relaxant responses. The same concentrations of PAMP had no effect on the aortic ring precontracted with phenylephrine. In the presence of d ifferent concentration of PAMP, ADM at 10 -8 mol·L -1 , however, did n o longer induce relaxant responses. Co-administration of ADM with ADT decreased the vascular tone. The percentage of decrease has no significant difference from that of administrating ADM alone. This effect of adrenomedullin was significant ly abrogated in the presence of PAMP. Conclusins These res ults demonstrated that ADM caused vasodilatation of isolated aortic ring constri cted with phenylephrine. The relaxing effect of ADM on the aortic ring can be an tagonized by PAMP, This effect of PAMP on ADM induced vasorelaxation, probably t hrough a NO-dependent pathway.
