摘 要:目的:研究淫羊藿苷(ICA)持续给药延缓兔椎间盘纤维环细胞退变的可能性。方法:分离培养兔椎间盘纤维环细胞并传至第1代,调整细胞密度后部分细胞作为P0组铺板后待测;剩余细胞分为空白组(15%FBS培养液培养)与ICA组(含ICA的15%FBS培养液培养,ICA浓度10-5M),该部分细胞持续培养至第3代时检测。通过MTT法检测细胞增殖、荧光定量PCR检测蛋白多糖表达、流式细胞仪检测细胞周期来评价淫羊藿苷对纤维环细胞生物学特性的影响。结果:与空白组比较,其余两组的细胞增殖、蛋白多糖表达、S期细胞比例均明显增高(P<0.05)。结论:第3代纤维环细胞已发生退变,淫羊藿苷持续给药可以延缓椎间盘纤维环细胞退变。Objective:To research the possibility of delaying annulus fibrous cells degeneration of icariin. Method:The annulus fibrous cells of rabbit were isolated and cultured. After adjusting the cell density of the first generation,some cells as the P0 group were decked to analysis,the others were divided into blank group(cultured with 15% FBS nutrient solution)and ICA group(cultured with 15% FBS nutrient solution with ICA,the ICA concentration is 10-5M). The cells of blank group and ICA group were cultured to the third generation continuously. The influence of icariin on biological characteristics of annulus fibrousus cells was evaluated by cell proliferation,the express of proteoglycan and cell cycle. Results:Compared with the blank group,cell proliferation,proteoglycan expression,proportion of S period cell of the rest two groups were increased(P<0.05).Conclusion:Third-generation annulus fibrosus cells have degenerated,icariin continue medicatine can delay the degeneration of annulus fibrous cells.
