柴胡疏肝散对肝郁大鼠血清NO和脑5-HT含量的影响

摘　　要：目的:探讨柴胡疏肝散对肝郁SD大鼠血清一氧化氮(NO)和脑中5-羟色胺(5-HT)含量的影响。方法:将大鼠随机分为空白组、模型组、治疗组3组,10只/组,雌雄兼用。采用鼠笼水平震荡、冰水游泳、拍打、悬尾、绳子捆绑四肢等方法对大鼠进行连续不定14 d刺激,建立肝郁证模型。治疗组用柴胡疏肝散对肝郁证模型大鼠进行治疗,空白组及模型组灌胃蒸馏水。ELISA试剂盒分别检测各组大鼠血清中NO和5-HT的含量。结果:与空白组比较,模型组大鼠脑5-HT含量升高(P<0.01),血清NO含量降低(P<0.05);与模型组比较,治疗组大鼠脑5-HT含量降低(P<0.05),血清NO含量升高(P<0.05)。结论:柴胡疏肝散能够升高肝郁SD大鼠血清NO水平和降低脑中5-HT的含量,此可能是其抗抑郁作用的机理之一。Objective:To study the effects of Chaihu Shugan powder on the contents of NO and 5-HT in the liver depression rats. Methods:Thirty SD rats,half male and half female,were divided into blank group,model group and treatment group.The rats were irritated by shaking the cages,letting them swim in icey water,flaping,suspending their tails,binding their limbs,etc,continuously and variably in 14 days to establish stagnation of liver qi models. The rats were treated with Chaihu Shugan powder in treatment group. The level of NO in serum and 5-HT in brain were detected. Results:Compared with the blank group,the level of 5-HT in brain was increased in the model group(P<0.01),the level of NO in serum was decreased in the model group(P<0.05). Compared with the model group,in the treatment group,the level of 5-HT in brain was decreased(P<0.05)and the level of NO in serum was increased(P<0.05). Conclusion:Chaihu Shugan powder can increase the level of NO in serum and reduce the content of 5-HT in brain of stagnation of liver qi model rats. This may be one of the anti-depressive mechanism of Chaihu Shgan powder.
