摘  要:目的:评价鹿红颗粒对心肌梗死后心力衰竭(CHF)模型大鼠血流动力学及血浆血管紧张素Ⅱ(AngⅡ)和醛固酮(ALD)的影响。方法:60只SD大鼠随机分配至模型组、假手术组、雅施达组及鹿红颗粒高、中、低剂量组,结扎左冠状动脉的方法制备心肌梗死后心力衰竭模型。药物干预8周后用电生理仪记录血流动力学变化,包括左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室内压最大上升和下降速率(±dp/dtmax);放射免疫分析法检测AngⅡ及ALD。结果:雅施达、鹿红颗粒高、中剂量均能有效升高LVSP、降低LVEDP、升高+d P/dtmax及-d P/dtmax(P<0.01),且鹿红颗粒高、中剂量组在改善血流动力学指标上优于雅施达组(P<0.01),而高、中剂量组之间无显著差异(P>0.05);雅施达与鹿红颗粒高、中剂量均能有效降低模型大鼠血浆AngⅡ和ALD的水平(P<0.01),而鹿红颗粒高、中剂量略差于雅施达(P<0.01),高、中剂量组之间无显著差异(P>0.05)。结论:鹿红颗粒能显著改善心力衰竭大鼠心脏血流动力学参数,降低心力衰竭大鼠血浆AngⅡ和ALD水平,从而改善心功能,延缓CHF病程进展。Objective: To observe the effect of 'Luhong Decoction'on hemodynamics and plasma angiotensin Ⅱ( AngⅡ) and aldosterone( ALD) in rats with heart failure. Methods: 60 heart failure rats were established by ligating left anterial descending coronary artery. They were randomly allocated into model group,sham operation group,perindopril group,Luhong low-dose,middle-dose and high-dose groups. 8 weeks after medicine administration,the rats from each group were detected by an electrophysiology system for hemodynamic changes including left ventricular systolic pressure( LVSP),left ventricular diastolic pressure( LVEDP),and the maximum rate left ventricular pressure rised or decreased( ± LVdp / dtmax),Ang Ⅱand ALD was detected by radio-immunity methods. Results: Perindopril and middle-dose,high-dose Luhong decoction can significantly improve LVSP, ± LVdp / dtmax and decrease LVEDP of model rats( P < 0. 01). Middle-dose and high-dose Luhong decoction were better than perindopril on improving the results said above( P < 0. 01). There was no difference between Luhong middle and high-dose groups( P > 0. 05). Perindopril and middle-dose,high-dose Luhong decoction can significantly lower Ang Ⅱ and ALD( P < 0. 01) and perindopril was better than Luhong on this effect( P < 0. 01). There was no difference between Luhong middle and high-dose groups( P > 0. 05). Conclusion: 'Luhong Decoction'can improve the hemodynamic performance of model rats and can lower plasma Ang Ⅱ and ALD of model rats which can improve heart function and delay the development of CHF.
