抗炎通管汤对慢性输卵管炎模型大鼠IL-2、IL-6及TNF-α表达的影响

摘　　要：目的:探讨抗炎通管汤对慢性输卵管炎模型大鼠输卵管组织中白介素-2(IL-2)、白介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)表达的影响。方法:60只大鼠随机分为正常组、模型组、金刚藤颗粒组及抗炎通管汤低、中、高剂量组,每组10只。除正常组外,其余各组大鼠采用"混合菌株接种法"制备慢性输卵管炎大鼠模型。造模后第11天开始灌胃给药,正常组和模型组大鼠每日灌胃注射用水2 ml,金刚藤颗粒组每日灌胃金刚藤颗粒药液2 ml(浓度为0.262 5 g/ml),抗炎通管汤低、中、高剂量组每日灌胃抗炎通管汤药液2 ml(给药浓度分别为9.375g/kg、18.75g/kg、37.5g/kg),连续用药20 d。采用免疫组化方法检测大鼠输卵管组织中IL-2、IL-6及TNF-α的表达。结果:与正常组比较,模型组及各给药组IL-2、IL-6及TNF-α表达均升高,差异具有统计学意义(P<0.05)。与模型组比较,金刚藤颗粒组和抗炎通管汤中、高剂量组IL-2表达下降,差异有统计学意义(P<0.05,P<0.01);抗炎通管汤高剂量组IL-6表达下降,差异有统计学意义(P<0.05);金刚藤颗粒组和抗炎通管汤高剂量组TNF-α表达下降,差异有统计学意义(P<0.05,P<0.01)。结论:抗炎通管汤可降低慢性输卵管炎模型大鼠输卵管组织中炎症相关因子的表达,且存在一定的量效关系。Objective: To study the effect of'Kangyan Tongguan Decoction'on the expressions of interleukin-2( IL-2),interleukin-6( IL-6) and tumor necrosis factor-α( TNF-α) in oviduct tissue of rats with chronic salpingitis. Methods: Sixty rats were randomized into the normal group,model group,'Jingangteng Granule'group and'Kangyan Tongguan Decoction'groups( low,medium and high dose),with 10 rats in each group. The rat models with chronic salpingitis were established by mixed bacteria inoculation method. From the 11 th day after modeling,the rats in the normal group and model group were treated with injection water of 2 ml,the rats in the 'Jingangteng Granule'group were treated with 'Jingangteng Granule'solution( 0. 262 5 g / ml) of 2 ml,and the rats in the 'Kangyan Tongguan Decoction'groups were treated with 'Kangyan Tongguan Decoction'( 9. 375 g / kg,18. 75 g / kg and 37. 5 g / kg respectively) of 2 ml,for 20 days continuously. The expressions of IL-2,IL-6 and TNF-α in oviduct tissue of rats were detected by immunohistochemical method. Results:Compared with the normal group,the expressions of IL-2,IL-6 and TNF-α in the model group and the administration groups were higher( P < 0. 05). Compared with the model group,the expression of IL-2 in the'Jingangteng Granule'group and the'Kangyan Tongguan Decoction'groups( medium and high dose) was lower( P < 0. 05,P < 0. 01); the expression of IL-6 in the'Kangyan Tongguan Decoction'group( high dose) was lower( P < 0. 05); the expression of TNF-α in the'Jingangteng Granule'group and the'Kangyan Tongguan Decoction'group( high dose) was lower( P < 0. 05,P < 0. 01). Conclusion:'Kangyan Tongguan Decoction'can reduce the expressions of inflammatory factors in oviduct tissue of rats with chronic salpingitis and there is some correlation between the quantity and the effect.
