摘 要:目的探讨苦参碱及联合顺铂对宫颈癌Si Ha细胞中TSLC1基因表达的影响。方法以人宫颈癌细胞株Si Ha细胞株为试验材料,采取甲基偶氮唑蓝法检测并计算细胞抑制率。采取实时荧光逆转录聚合酶联反应法分析TSLC1 mRNA基因表达情况。结果顺铂、苦参碱联合顺铂均可显著抑制Si Ha细胞体外增殖,差异具有统计学意义(P<0.05);随着用药浓度的增加,抑制作用增强,不同浓度两两比较,差异具有统计学意义(P<0.05);联合组细胞抑制率高于苦参组、顺铂组,差异具有统计学意义(P<0.05)。三组用药后,TSLC1基因表达水平均高于对照组,差异具有统计学意义(P<0.05);随着浓度的增加,影响效果更强,组间不同浓度两两比较,差异具有统计学意义(P<0.05);联合组高于顺铂组、苦参碱组,同浓度比较,差异具有统计学意义(P<0.05);顺铂组与苦参碱组同浓度比较,差异无统计学意义(P>0.05)。结论苦参碱及联合顺铂对宫颈癌Si Ha细胞中TSLC1基因表达有抑制作用,且与药物浓度呈剂量依赖性。Objective To explore the effects of matrine combined with cisplatin on the expression of TSLC1 gene in human cervical cancer Si Ha cell. Methods Human cervical cancer Si Ha cell were selected as the experimental material,the inhibition rate was tested by MTT,the expression of TSLC1 gene were tested by RT-PCR. Results The matrine combined with cisplatin can inhibit proliferation of Si Ha cell in vitro( P < 0. 05); The inhibitory effect in high concentration was higher than that of the lower concentration( P < 0. 05); The inhibition rate of the combined group was higher than matrine group and cisplatin group( P <0. 05); The TSLC1 gene expression of the 3 groups was higher than that of the control group( P < 0. 05); The inhibitory effect in high concentration were higher than that of the lower concentration( P < 0. 05); The combined group was higher than the matrine group and cisplatin group( P < 0. 05); There was no significant difference between matrine group and cisplatin group( P > 0. 05).Conclusion Matrine combined with cisplatin has inhibitory effect on the expression of TSLC1 gene in human cervical cancer SiHa Cell,and the effect is dose dependent.
