摘 要:目的利用建立的Taq荧光定量RT-PCR检测方法研究鸭肝炎病毒(DHV-1)强毒株在感染雏鸭体内早期的动态分布规律。方法本研究采用鸭肝炎病毒强毒株人工感染3日龄雏鸭,利用已建立的Taq荧光定量RT-PCR方法,对接种24 h内DHV-1强毒株感染雏鸭的组织脏器进行了定量检测。结果接种后4 h即可在雏鸭心、肝、脑、肺、胰腺、回肠、盲肠、直肠、法氏囊和脾脏组织中检测到一定水平的病毒RNA,随着病程发展,RNA拷贝数持续升高,接种24 h后,肝脏中的病毒RNA拷贝数为最高,达到108.81copies/g,心、肺、肾、脾次之,约107copies/g。结论DHV-1强毒在雏鸭的早期感染雏鸭的实质器官、免疫器官和消化系统等均具有广泛的嗜性。Objective To investigate the dynamic distribution of duck challenge hepatitis virus strain( 161 /79 / V)in experimentally infected ducklings using the Taqman real-time fluorescent quantitative PCR. Method Ducklings were infected with the 161 / 79 / V strain at 3-day age,and the virus RNAs were extracted from different organs within 24 hours and quantified using the fluorescent quantitative PCR method. Result DHV RNAs were detected in heart,liver,brain,lung,pancreas,ileum,cecum,rectum,bursa and spleen 4 hours after inoculation,and the RNA copies continuously increased. The highest copy of the DHV RNA was detected in liver( 108. 81 copies / g) 24 h after inoculation,and the RNA copies were somewhat low in heart,lung,kidney and spleen( about 107 copies / g).Conclusion Our study indicates that the challenge DHV strain 161 / 79 / V is tissue or organ ecotropic,such as the solid organ,the immune organs and the digestive system of the infected ducklings.
