摘 要:目的 :为探讨妇科抗肿瘤新方法 ,检测单用维生素K3或与 4Gyγ 射线照射联合应用对人宫颈癌HeLa细胞细胞毒性的影响。方法 :水溶性维生素K3处理 2 4h ,合用或不用γ 射线照射人宫颈癌HeLa细胞 ,应用碱性彗星试验 (alkalinecometassay) ,也称碱性单细胞凝胶电泳法 ,测定彗星尾矩和彗星长度。结果 :对彗星尾矩影响 :单用维生素K31 0 ,2 0 ,40mmol·L- 1 ,彗星尾矩为空白对照组的 2 ,3和 5倍 ;与 4Gyγ 射线照射合用组是单纯用药组的 2 .5~ 4倍 ,差异有显著的统计学意义 (P <0 .0 1 )。实验组间比较 ,4Gyγ 射线照射与维生素K3合用组 1 0与 40mmol·L- 1组间有差异。对彗星长度影响 :单纯用药培养 2 4h ,各剂量组均比对照组的彗星长度值明显增加 (P <0 .0 1 ) ;4Gyγ 射线与维生素K3合用 ,实验组与对照组比较差异有显著统计学意义 (P <0 .0 1 ) ;合用组的 1 0与 40mmol·L- 1 ,2 0与 40mmol·L- 1 组间存在差异。结论 :维生素K3对人宫颈癌HeLa细胞产生细胞毒性 ,与γAIM: To explore cytotoxicity induced by γ radiation and menadione sodium bisulfates on HeLa cells in order to develop a new method for anticancer in gynecological oncology. METHODS: Alkaline comet assay was used to test tail moment and comet length of the cell line after treatment with menadione or a combination of 4Gy γ radiation with the drug. RESULTS: The values of tail moment of the cells treated only with 10, 20, and 40 mmol·L -1 of menadione were 2, 3, and 5 times as high as that of the control without treatment (P< 0.01 ). The values of tail moment after treatment with a combination of 4 Gy γ radiation with menadione were 2.5 -4 times as high as that only with the menadione (P< 0.01 ). Differences were significant (P< 0.01 ) between groups treated with 4Gy γ radiation and 10, and 40 mmol·L -1 of menadione, and the groups incubated only with menadione. Compared with the control, comet length increased significantly in groups incubated only with menadione. Differences were significant between the groups incubated with the drug in concentrations of 10 and 40 mmol·L -1 , as well as 20 and 40 mmol·L -1 . Treatment of 4Gy γ radiation combined with menadione significantly increased the comet length in each group, as compared to that treated only with meadione. Differences were significance between groups incubated with the drug in concentrations of 10 and 40 mmol·L -1 , 20 and 40 mmol·L -1 . CONCLUSION: Menadione can enhance the cytotoxicity to HeLa cells in combination with γ radiotherapy.
