摘 要:目的 研究年龄对中国成年男性血清褪黑素水平及其昼夜节律性的影响。方法 健康志愿者按年龄分为四组 :30~ 39岁组 (8例 )、40~ 49岁组 (5例 )、50~ 59岁组 (5例 )和≥ 60岁组 (7例 )。白天 (8~ 2 2时 )每 2 h、夜间 (2 2~ 8时 )每 1 h采血一次 ,放免法检测血清褪黑素浓度。采用 t检验、ANOVA及余弦法分析处理观测数据。结果 血清褪黑素水平呈明显的昼夜节律性变化 ,高峰在午夜 ,白天水平很低 ,采样时间对血清褪黑素水平具有非常显著性影响 (P<0 .0 1 )。余弦法血清 MT仅在 30~ 39岁和 40~ 49岁组中有显著昼夜节律性变化 (P<0 .0 5)。 30~ 50岁组血清褪黑素水平相似 ,≥ 60岁组夜间 (2 3~ 5时 )血清褪黑素水平明显降低 ,与 30~ 50岁组相比有非常显著性差异 (P<0 .0 1 )。结论 血清褪黑素水平及其昼夜节律性变化受年龄的影响 ,随年龄增长夜间血清褪黑素水平下降 ,并且其昼夜节律性 (50~ 59岁和≥ 60岁组 )消失。Objective Melatonin, the hormone produced by pineal gland during the dark phase of circadian cycle, has been proposed to be involved in age process and age related diseases. In order to investigate influences of age on serum melatonin, we examined circadian variations of serum melatonin concentrations in 25 healthy Chinese men age from 31 to 63 years old.Methods The subjects were divided into four age groups, i.e., 30s (N=8), 40s (N=5), 50s ( n =5), and 60s( n =7). Serum samples were drawn at 2 h intervals from 8:00 to 22:00 and hourly from 22:00 to 8:00 for a total of 24 hours. RIA has been used to determine serum melatonin.Results Data were analyzed by Student t test and ANOVA, the circadian rhythmicity was analyzed also by cosinor method.Our results showed circadian serum variations of melatonin levels with a peak at night, very low levels during daytime and with similar general patterns in all age groups. However, a statistically significant circadian rhythm ( P < 0.05) was validated only in the younger groups (i.e., 30s and 40s). No significant difference was found between 30s, 40s, 50s, while nocturnal melatonin level(23:00~05:00)in 60 s was significantly lower ( P < 0.01) than that in other younger age groups (30~50 s).Conclusions Our results suggest that aging influenced on both nocturnal serum melatonin level and its circadian rhythmicity.
