摘  要:目的研究葡萄糖(GLU)、胰岛素(INS)对内皮细胞(ECs)一氧化氮 (NO)和内皮素 -1(ET-1)生成及其mRNA水平的影响 ,探讨糖尿病血管并发症与血管内皮功能异常的关系。方法用放免法测定ECscGMP和ET-1水平,cGMP用来反映NO的量 ;半定量RT-PCR检测一氧化氮合酶 (eNOS)mRNA和ET-1mRNA水平。结果 (1)高浓度GLU(20mM,40mM)能促进ECs合成cGMP和ET-1 ,并上调eNOSmRNA水平;(2)INS(0.18nmol/L~6nmol/L)能促进ECs生成cGMP ,Ins(1.8nmol/L~6nmol/L)能增加ECs合成ET -1 ,并上调ET-1mRNA水平 ;(3)在高浓度GLU下 ,INS(1.8nmol/L)刺激NO的生成作用明显降低。 (4)在相同浓度GLU(20mM,40mM)、INS(1.8nmol/L~6nmol/L)下 ,ET-1增加的倍数远高于cGMP增加的倍数。结论高浓度GLU和INS可直接导致血管内皮细胞功能的异常,促进了糖尿病血管并发症的发生、发展。Objective To explore the relationship between diabetic vascular complications and the disfunc-tions of vascular endothelial cells(ECs),the effects of various concentrations of glucose and insulin on nitric oxide(NO)and endothelin-1(ET-1)production and the levels of their mRNA of vascular endothelial cells in culture were explored.Methods cGMP(as an indicator of NO production)and ET-1radioimmunoassay for ECs were used in this study,and the levels of eNOS mRNA and ET-1mRNAwere measured by semiquantiative RT-PCR.Results(1)High concentrations of glucose(20mM~40mM)increased cGMP and ET-1release of ECs,and upregulated the level of eNOS mRNA.(2)Insulin(0.18nmol/L~6.0nmol/L)increased cGMP release of ECs;Insulin(1.8nmol/L~6.0nmol/L)increased release ET-1,and upregulated the level of ET-1mRNA.(3)Under high concentrations of glucose,stimulation of cGMP levels by insulin was significantly decreased.(4)Under same con-centrations of glucose(20mM-40mM)and insulin(1.8nmol/L~6.0nmol/L),the increasing multiples of ET-1were much more than those of cGMP.Conclusions High concentrations of glucose and insulin can lead to dys-function of vascular endothelial cells directly,which accelerates the development of diabetic vascular complications.
