摘　　要：目的　通过检测组织因子mRNA在异种心脏移植急性血管排斥反应期的动态表达与排异反应强度之间的关系 ,初步了解组织因子在该反应期的意义。方法　建立豚鼠 大鼠异种异位心脏移植动物模型 ,使用CVF抑制超急性排异反应 ,FK5 0 6抑制细胞免疫排斥反应 ,分别在移植术后 4、8、12、16、2 4h切取移植心脏 ,通过病理切片观察排异反应和凝血强度 ,同时通过RT PCR方法检测组织因子mRNA表达强度 ,未移植豚鼠心脏作对照。结果　在移植后 ,移植心脏的排异反应和凝血逐渐加重 ,豚鼠组织因子mRNA表达强度逐渐减弱 ,大鼠组织因子mRNA表达强度逐渐增强。结论　组织因子在异种心脏移植急性血管排斥反应期中起重要作用 ,其中供体的组织因子可能与凝血的激发有关 ,受体的组织因子可能与凝血的发展有关。Purpose: To evaluate the significance of tissue factor in acute vascular rejection of xenotrantation. Methods: We established the model of acute vascular rejection of heart xenotransplantation. Gurinea pigs and SD rats were served as donors and recipients respectively. Cobra venom factor (CVF) was given to prevent hyperacute rejection and FK506 was given to prevent cellular rejection. The xenograft recipients were killed and cardiac xenografts harvested 4, 8, 12, 16, 24 hours after transplantation. Histologic examination was used to demostrate the degree of rejection and coagulation while revers transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of tissue factor mRNA, and the guinea pigs hearts, harvested but not transplanted, were studied as controls in all samples. Results: During the period of post-transplantation, the blood coagulation in the cardiac graft became more and more serious. The expression of guinea pig tissue factor mRNA decreased while the expression of SD rat tissue factor mRNA increased. Conclusions: The tissue factor act as an important role in the acute vascular rejection of heart xenotransplantation. These results suggest that in acute vascular rejection, coagulation is initated by the donor tissue factor and developed with the effect of the recipient tissue factor.
